乙醛脱氢酶2基因多态性与脑梗死相关性研究

目的探讨乙醛脱氢酶2（ALDH2）基因多态性与脑梗死的相关性。方法以156例经MRI确诊的脑梗死患者为脑梗死组，以同期MRI排除脑梗死诊断的160例患者为对照组，取外周血标本提取DNA，应用聚合酶链式反应技术扩增目的基因片段，分析ALDH2基因型与脑梗死的关系。结果单因素分析，AA基因型脑梗死的危险性是GG基因型的4．13倍（P〈0．01）。多元Logistic回归分析，A等位基因脑梗死的危险型是G等位基因的2．35倍（P〈0．01）。结论ALDH2基因G487A多态性与脑梗死的发病有关，A等位基因增加脑梗死的危险性。

THE RELATIONSHIP BETWEEN GENETIC POLYMORPHISM OF ALDEHYDE DEHYDROGENASE - 2 AND CEREBRAL INFARCTION

Objective To investigate the correlation between aldehyde dehydmgenase- 2 （ALDH2） gene polymorphisms and cerebral infarction（CI）. Methods A case- control study of 156 patients with MRI documented CI and 160 control subjects with a normal MRI was conducted. Genotype was determinde by polymerase chain reaction- restriction fragment length polymorphism（PCR - RFLP） for the common G487A functional promoter polymorphism of the ALDH2 gene. Fatalness of genetic tactors toward CI was analyzed by Logistic regression. Results The frequencies of ALDH2 ＊ 2/＊ 2 and ALDH2 ＊ 1/＊ 2 genotypes in case group group were higher than that in the control group（P〈0.01）. Multivarate Logistic regression analysis showed that A allele, hypertension and diabetes were risk factors for cerebral infarction. Conclusion The G487A polymorphism is significantly associated with CI, and allele A variant of aldehyde dehydmgenase- 2（ALDH2）gene polymorphism may be a risk facor for cerebral infarction.
