脐血T淋巴细胞亚群分布特点及其临床意义探讨

目的比较脐血、骨髓及外周血中T淋巴细胞亚群的分布特点,探讨脐血移植后免疫重建延迟的发生机制.方法应用流式细胞仪分别检测脐血、骨髓及外周血中CD3 、CD3 CD5 、CD4 、CD4 CD28 、CD8 和CD8 CD28 细胞的含量.结果 CD3 、CD4 及CD8 细胞数在脐血、骨髓及外周血中无显著差异,P> 0.05.脐血中CD3 CD5 、CD4 CD28 及CD8 CD28 细胞数显著低于骨髓及外周血,P<0.05.骨髓与外周血中各T淋巴细胞亚群测定无显著差异,P>0.05.结论脐血中成熟的T淋巴细胞(CD3 CD5 )及细胞毒T淋巴细胞(CD4 CD28 、CD8 CD28 )的数量显著低于骨髓及外周血,这可能是脐血移植后免疫重建延迟的主要原因.

Distributive characteristics and clinical significance of T lymphocyte subsets in cord blood

Objective To study the mechanism of delayed immune reconstitution after cord blood transplantation by compared with the characteristics of distribution of T lymphocyte subsets in cord blood,bone marrow and peripheral blood.Methods The amounts of CD3 ,CD3 CD5 ,CD4 ,CD4 CD28 ,CD8 and CD8 CD28 cells in cord blood,bone marrow and peripheral blood were respectively measured with flow cytometry.Results There was no marked change on the amounts of CD3 ,CD4 and CD8 cells in cord blood,bone marrow and peripheral blood( P >0.05 ).The amounts of CD3 CD5 ,CD4 CD28 and CD8 CD28 cells in cord blood were significantly decreased compared with bone marrow and peripheral blood( P <0.05 ).The amounts of each T lymphocyte subsets in bone marrow were not significantly changed compared with peripheral blood( P >0.05 ).Conclusion The amounts of mature T lymphocyte(CD3 CD5 ) and cytotoxic T lymphocyte(CD4 CD28 ,CD8 CD28 ) in cord blood were significantly decreased compared with bone marrow and peripheral blood.Maybe this is the main cause of delayed immune reconstitution after cord blood transplantation.
