疏肝理气方剂对大鼠胃窦组织神经元及神经递质的影响

摘    要:目的观察疏肝理气方剂对一氧化氮合酶(NOS)阳性神经元、一氧化氮(NO)和乙酰胆碱(Ach)神经递质的影响,从而探讨疏肝理气方剂促胃动力作用及其机制。方法将SD大鼠随机分为小柴胡汤组、四逆散组、柴胡疏肝散组、解肝煎组、逍遥散组、莫沙比利阳性对照组及生理盐水空白对照组。采用NADPH-黄递酶组化法染色显示大鼠胃窦部NOS阳性神经元,医学彩色图像分析仪图像定量分析;采用放射免疫法检测大鼠用药后胃窦组织中NO、Ach含量。结果小柴胡汤组、四逆散组、柴胡疏肝散组大鼠胃窦黏膜肌内NOS阳性神经元的面积及平均吸光度值明显降低,较空白对照组具有显著性差异(P<0.05或P<0.01)。小柴胡汤组、四逆散组及柴胡疏肝散组胃窦组织中NO含量明显减少,Ach含量增多,与空白对照组均有显著性差异(P<0.05或P<0.01)。结论疏肝理气方剂中,小柴胡汤、四逆散、柴胡疏肝散可显著减少大鼠胃窦组织中NOS阳性神经元,降低NO含量,并同时增加Ach的含量,从而促进胃肠运动,缓解胃肠动力障碍。

Abstract:ObjectiveTo observe the effects of five ancient herbal formulae on nitric oxidesynthase(NOS),NO and Ach for further research on its mechanism. Methods SD rats were randomized into seven groups:Xiaochaihu group,Sini group,Chaihu Shugan group,Jiegan group,Xiaoyao group,control group and normal group. NADPH-diaphorase histochemistry dye was abopted to display NOS neurons in gastric antrum,and radioimmunoassay was used to detect the comtents of NO and Ach in gastric antrum. Result The XiaoChaihui group,Sini group and Chaohu Shugan group could obviously decrease NOS positive nerve fiber,ending areas as well as light density;with significant differences in comparison with normal group (P<0.05,P<0.01);on the other hand,these three groups could reduce NO content and increase Ach comtent,with significant differences in comparison with normal group(P<0.05,P<0.01). Conclusion These three formulae can reduce NOS positive neuron and NO content and increase Ach to promote stomach movement and reliveve gastric kinestic disorder.
