胃癌中COX-2基因的表达及其与Bcl-2和CyclinD1相关性研究

目的 研究胃癌中环氧合酶-2（cox-2）的表达情况和生物学意义，以及与Bcl-2和CyclinDl的相关性。方法 选择胃癌42例和浅表性胃炎24例，对COX-2、Bcl-2和Cyclin D1抗体行SP法免疫组化染色。结果42例胃癌标本中有31例COX-2阳性，24例中浅表性胃炎中5例弱阳性表达（P〈0．01）。CyclinD1和Bcl-2在胃癌中的阳性率分别为47．6％（20／42），50％（21／42）。COX-2和CyclinDl、Bcl-2表达呈正相关。Cox-2基因表达与年龄、性别、大小、组织学类型和浸润深度无关。但与肿瘤的淋巴结转移具有显著的相关性（P〈O．05）。结论 COX-2过表达可能通过对CyclinDl和Bcl-2表达的调控。抑制胃癌细胞凋亡，加速细胞增殖，促进胃癌的发生发展；COX-2过表达在肿瘤侵袭和转移过程中发挥着重要作用。

Expression of COX- in Gastric Carcinomas and Its Correlation with Bcl-2 and CyclinD1

Objective To explore the biological significances of COX-2 in patients with gastric cancer and its correlation with bcl-2 and cyclinD1.Methods Forty-two cases of gastric carcinomas and 24 superficial gastritis were detected for COX-2,Bcl-2 and CyclinD1 with immunohistochemistry.Results Among 42 gastric carcinomas,31 showed COX-2 positive,while 5 cases did the positive in 24 normal mucosas(P<0.01).The positive rates of Cyclin D1 and Bcl-2 in cancer and gastritis were 47.6 %(20/42) and 50%(21/42),respectively.The expression of COX-2 was positively correlated with those of CyclinD1 and Bcl-2(P<(0.05)).COX-2 expression was not relative with such clinicopathological parameters as age,sex,size,histological types.However,there was a positive relationship between COX-2 gene expression and lymph node metastasis.Conclusion The study demonstrated that the overexpression of COX-2 gene may participate in oncogenesis and progression of gastric carcinoma by inducing high expression of Bcl-2 and Cyclin D1;COX-2 plays an important role in the process of invasion and metastasis of tumor cells.
