不同治法对急性肺损伤小鼠核转录因子表达的影响

摘    要:目的:探讨开达膜原法、清热解毒法、清热凉血法对急性肺损伤小鼠核转录因子(NF-κB p50)表达的影响。方法:采用小鼠尾静脉注射LPS(10 mg/kg)建立小鼠急性肺损伤模型,将小鼠随机分为正常对照组、模型对照组、清热解毒组、清热凉血组及开达膜原组。观察模型对照组与清热解毒组、清热凉血组、开达膜原组NF-κBp50水平的变化情况、肺体指数及肺组织病理学形态。结果:病理学观察,模型对照组肺组织呈大片出血、坏死,大量炎细胞浸润;清热解毒组局部肺组织中仍见小出血灶,肺组织结构未被破坏,局部细支气管细胞增生(轻度);清热凉血组肺组织见一处小灶性肺出血灶;开达膜原组见轻度肺间质炎症。与模型对照组相比,清热解毒组小鼠肺体指数明显降低(P〈0.05);开达膜原组、清热解毒组、清热凉血组小鼠NF-κBp50阳性细胞率均降低(P〈0.05~0.01)。结论:清热解毒法、清热凉血法及开达膜原法均对急性肺损伤小鼠肺组织有保护作用,其机制可能与抑制NF-κBp50的表达有关。

Abstract:Objective:To approach the effects of eliminating pathogenic factor from Moyuan therapy,heat-clearing and toxin-relieving therapy,and removing heat to cool blood therapy on nuclear factor-kappa B p50 in acute lung injury(ALI).Methods:The model of ALI in mouse was established by intravenous injection of lipopolysaccharide(10 mg/kg).Mice were randomized into normal control group,model control group,heat-clearing and toxin-relieving group,removing heat to cool blood group,and eliminating pathogenic factor from Moyuan group.Levels of nuclear factor-kappa B p50,lung-body index,and histopathologic changes in lung tissue were observed.Results:There were massive bleeding,necrosis,and inflammatory cell infiltration in lung tissues of model control group;in heat-clearing and detoxicating group,there were mild bleeding,bronchiolar epithelial hyperplasia in local lung tissues without destruction of the structure of lung tissue;there was a small hemorrhagic focus in lung tissue of removing heat to cool blood group,and mild inflammation in lung interstitium of eliminating pathogenic factor from Moyuan group.Compared with normal control group,the lung-body index in heart-clearing and toxin-nelieving group was higher,P<0.05.Compared with model control group,the positive ratios of nuclear factor-kappa B p50 in three treatment groups were significantly lower,P<0.05-0.01.Conclusion:Heat-clearing and toxin-relieving therapy,removing heat to cool blood therapy and eliminating pathogenic factor from Moyuan therapy can all protect lung tissue in ALI,which may be associated with the suppression of the expression of nuclear factor-kappa B p50.
