摘    要:目的研究不同冷冻载体对小鼠卵巢组织玻璃化冻存的影响.方法将9~12周龄小鼠的卵巢组织取出,在冷冻液中平衡后,分别以闭合麦管、开放麦管与冷冻环为冷冻载体快速深低温冻存,解冻后行自体双肾被膜下移植,观察动情周期、移植21 d后取存活移植物固定,观察单位高倍视野下卵泡数、测定小鼠血清雌激素水平,对比冻存效果.结果移植后各组均有移植物存活,移植物内可见各级处于不同发育阶段的卵泡.闭合麦管、开放麦管与冷冻环组的移植物存活率分别为78.5%、78.5%、84.6%;单位高倍镜下的卵泡数分别为4.0±0.8、4.5±0.7、4.6±0.9;小鼠血清雌激素水平分别为(10.4±0.8)pg/mL、(11.0±1.6)pg/mL、(12.5±1.1)pg/mL.其中,冷冻环组在移植物存活率及小鼠血清雌激素水平与闭合麦管组相比存在显著性差异.结论采用冷冻环做载体能明显提高冻融移植后卵巢组织的存活率与雌激素分泌功能,明显提高对小鼠卵巢组织的保存效果.

Abstract:Objective To observe the effect of different carriers on mouse ovarian tissue vitrification.Methods Ovaries tissue slices from mouse about 9~12 weeks were vitrified by close straw,open straw or cryoloop.After thawed,autotransplantations were performed and two pieces of ovarian tissue slices were respectively inserted under the kidney capsule of two sides in each mouse.The survival rate of grafts,histological structure,follicular density and serum E2 level in each group were detected after autotransplantation for 21 days.The effect of different freezing carriers on viability of frozen-thawed ovarian tissue was compared.Results The survival rate of grafts after 21 days autotransplantation in close straw group,open straw group or cryoloop group was respectively 78.5%,78.5% and 84.6%;the number of follicles in per high power field was 4.0±0.8,4.5±0.7and 4.6±0.9;the serum E2 level of mouse was 10.4±0.8 pg/mL,11.0±1.6 pg/mL and 12.5±1.1 pg/mL.Among them,there was significant difference between cryoloop group and close straw group in the survival rate of graft and the serum E2 level(P〈0.05).Conclusions The survival rate of graft and estradiol secretion can be improved by using cryoloop as vitrification carrier.
