| 摘 要： | 目的：探讨建立人角膜缘干细胞系，为组织工程研究提供足够的细胞储备。方法：取健康人角膜缘深部色素区组织块，经消化后，在含有RPMI—1640、20％胎牛血清的培养瓶和以羊膜细胞外基质为培养载体的培养皿中分别进行体外培养，并对连续传代的细胞行光镜、扫描电镜观察，比较细胞形态的变化并进行细胞计数与冻存处理。结果：人角膜缘干细胞具有人上皮细胞培养的特性，在培养液中传33代后，细胞仍然保持旺盛的分化、增殖能力，并且更适宜在羊膜细胞外基质上生长；目前我们冻存细胞的复苏率为82．2％。结论：将人角膜缘干细胞经体外培养33代并逐代冻存后初步建立了人角膜缘干细胞系。 |

| Abstract: | Objective To establish and reserve human limbal stem cell lines.Methods:Two digestive human limbal tissues were cultured in RPMI-1640 containing 20% fatal bovine serum.After 3 days,the human limbal stem cells cultured in two mediums.One in cultured glasses,the other on amniotic extracellular matrix(AECM).To observe the configuration and vitality,all of them were frozened photographed and examined by light microscope (LM),electron microscope (EM).Results:The cultured human stem cells had characteristics of epitheliums.AECM was an idea carrier in the limbal stem cells cultured.After passaged 33,the stem cells still remained the potency of proliferation and differentiation.By now,the rate of frozen cells' anabiosis was 82.2%.Conclusion:The human limbal stem cell lines have been preliminarily established. |
