自制中药颗粒对铅中毒模型大鼠学习记忆的 干预作用研究

摘    要:摘要:目的 探讨慢性铅中毒导致的模型大鼠学习记忆能力损伤及中药的干预作用。方法 60只大鼠随机分为2组,其中空白组12只,饮用双蒸水;造模组48只,饮含0.02%醋酸铅的双蒸水,连续60 d后将造模动物随机分为4组:高剂量治疗组(按3.0 g·kg-1·d-1的剂量灌胃自制中药颗粒)、低剂量治疗组(0.6 g·kg-1·d-1)、阳性对照组(依地酸钠钙加普鲁卡因肌肉内注射,50 mg·kg-1·d-1)及模型组(不予任何治疗),连续60 d后进行隐藏站台试验及空间探索试验,采血检测血铅,RT-PCR检测脑组织海马区NR1、NR2B mRNA表达。结果 经统计,与模型组比较,其他各组体重、血色素、穿过平台次数、第3象限活动时间、NR1、NR2B mRNA表达均显著增加(P<0.05),血铅含量、Morris水迷宫试验d 4潜伏期显著降低(P<0.05);与阳性对照组比较,中药高剂量组体重、第3象限活动时间、NR1 mRNA表达、NR2B mRNA表达显著增加(P<0.05)。结论 自制中药颗粒通过增加脑组织海马区NR1 mRNA、NR2B mRNA的表达,改善慢性铅中毒导致的学习记忆能力损伤。

Abstract:Abstract: Objective This study was to investigate the intervention effect of Chinese herbs on leaning-memory damage in rat hippocampus resulted from lead poisoning. Methods 60 SD rats were divided into 2 groups: 12 in the control group and others were in the model group. Chronic lead poisoning model were built by feeding with 0.02% lead acetate water for 60 days. Then these models were randomly divided into 4 groups: high-dosage group, low-dosage group, EDTA group and pathological group. The groups were then irrigated with the corresponding medicine. After 60 days, Morris Water maze test was conducted to the rats. Blood lead level was measured by atomic absorption spectrometry, and RT-PCR was used to determine the expression of N-methl-D- aspartate receptor 1 (NR1) and N-methl-D-aspartate receptor 2B (NR2B). Results Compared with the model group, significant increase in body weight, hemoglobin, the number of pass through the platform, the active time of the third quadrant and the expression of NR1 and NR2B was observed in other groups (P<0.05). Compared with the EDTA group, the weight, the active time of the third quadrant and the expression of NR1, NR2B of the high-dosage group were significantly higher (P<0.05). Conclusion Lead may induce rat memory damage by lowering the expression of NR1 and NR2B. TCM would reduce the damage.
