摘    要:摘要:目的 建立检测志贺菌CRISPR相关基因的斑点杂交方法。方法 分别采用PCR 法制备地高辛标记DNA 探针和人工合成的寡核苷酸探针,与志贺菌CRISPR相关基因进行斑点杂交。结果 设计出志贺菌CRISPR的相关基因探针;实现了志贺菌CRISPR系统的探针制备及杂交方法的建立。结论 斑点杂交可以快速大量检测志贺菌CRISPR的相关基因。

Abstract:Abstract: Objective This work was to establish a dot-blot hybridization for the detection of clustered regularly interspaced short palindromic repeat (CRISPR) with DNA probe in Shigella. Methods PCR was carried out to prepare the digoxingenin (DIG) -labeled DNA probe and oligonucleotide probe to detect CRISPR associated genes in Shigella by dot blot hybridization. Results Probes for CRISPR associated genes in Shigella were designed successfully. The establishment of probe and hybridization of Shigella CRISPR system was completed. Conclusion Dot blot hybridization is a rapid and sensitive method for the detection of CRISPR in Shigella.
