摘    要:摘要:目的 观察不同性别和年龄的健康人群外周血单个核细胞(PBMC)中Toll样受体3(TLR3)和Toll样受体7(TLR7)mRNA表达,探讨性别和年龄对TLR表达的影响。方法 选择健康人群113名,取抗凝全血,实时荧光定量PCR检测TLR3和TLR7 mRNA表达。结果 健康青年女性TLR3表达量比中老年女性高,差异有统计学意义(t=34.65,P<0.05),而TLR7基因表达水平的差异无统计学意义(t=1.72,P>0.05),健康青年男性TLR3,TLR7表达量比中老年男性高,差异均有统计学意义(t=17.63,7.17,P<0.05)。健康中老年女性TLR7表达量比中老年男性高,差异有统计学意义(t=4.03,P<0.05),但是TLR3表达量的差异没有统计学意义(t=-0.41,P>0.05)。健康青年女性TLR3,TLR7表达量比青年男性高,差异均有统计学意义(t=20.57,11.66,P<0.05)。结论 性别和年龄与对Toll样受体的表达有一定关系,为疾病预防和临床个体化治疗提供新的思路。

Abstract:Abstract: Objective The study aimed to investigate the expression of TLR3 and TLR7 mRNA in peripheral blood mononuclear cells (PBMC) of patients in healthy individuals, and thus to explore the effects of gender and age on TLR expression. Methods Peripheral blood was collected from 113 healthy individuals. Expression levels of TLR3 and TLR7 mRNA were detected by real-time quantitative PCR. Results The expression of TLR3 was significantly higher in young females than mid and old aged females (t=34.65, P<0.05). However, the expression of TLR7 gene was not significantly different between young females than mid and old females (t=1.72, P>0.05). The expression of TLR3 and TLR7 mRNA were significantly higher in young male group than mid and old male (t=17.63, 7.17, P<0.05), respectively. The expression of TLR7 was significantly higher in mid and old females than mid and old males (t=4.03, P<0.05). However, the expression of TLR3 gene was not significantly different between mid and old females than mid and old males (t=-0.41, P>0.05). The expression of TLR3 and TLR7 mRNA were significantly higher in young female group than young male, respectively (t=20.57, 11.66, P<0.05). Conclusion Gender and age discrepancies have a relationship with the expression of Toll-like receptors, which may provide new ideas for disease prevention and clinical individual therapy.
