大鼠PDLLA/NGF套管模型与自体神经移植模型脊髓背角c-fos表达的比较研究

摘    要:目的建立大鼠PDLLA/NGF(外消旋聚乳酸/神经生长因子)复合套管模型和大鼠外周神经自体神经移植神经再生模型,观察对比各模型神经恢复过程中机械痛觉超敏的变化及脊髓背角c-fos表达的改变,探讨两种神经再生模型中神经再生与神经性疼痛的联系。方法雄性Wistar大鼠60只随机分成3组,A组为自体神经移植再生模型组;B组为PDLLA/NGF套管修复模型组;C组假手术组。分别制成模型后,术后18d起检测机械刺激阈值及c-fos表达计数。所有数据采用均值±标准差(x±s)表示,组间采用t检验,用SPSS11.0软件进行统计分析,P<0.05为有统计学差异。结果A、B组各项指标与C组比较有显著差异;A组与B组比较c-Fos表达阳性细胞计数及机械痛阈无显著差异。结论两种模型再生神经生长进入去神经支配区域时,均出现支配区域的痛觉超敏和疼痛相关行为,神经性疼痛程度无差异。

Abstract:Objective To observe mechnic allodynia and c-fos expression changes between PDLLA/NGF compound condit model and nerve autograft model after models of rat were built,and study the corelation between two nerve regeneration model and neuropathic pain.Methods60 male wistar rats were randomly divided into four groups:sciatic nerve autograft(group A,n=20)PDLLA/NGF compound condit group(group B,n=20)and sham-operation(group C,n=20).After the model were built,on the 18d,we tested sensitivity threshold and c-fos counting.All data were described as mean and standard deviation(x±s),tested by means of T test,and statistically analyzed by SPSS11.0 software.P<0.05 is set as having statistical significance.ResultsCompared with group C,all indexes in group A and B have significant difference;Compared with group B,sensitivity threshold and c-fos counting in group A have no significant difference.ConclusionWhen the injured fibers have regrown into the nerve-yield region,both two model of nerve regeneration produced allodynia and pain-related behavior.There is no significant difference in the degree of neuropathic pain.
