X射线对人晶状体上皮细胞增生的影响

摘    要:目的研究X射线照射对人晶状体上皮细胞增生的影响.方法取对数增生状态的人晶状体上皮细胞,分别以50 cGy、100 cGy、200 cGy、300 cGy、400 cGy、500 cGy X射线照射,观察照射前后细胞形态学改变;MTT法测定X射线对细胞增生的影响;采用荧光标记、流式细胞仪测定增生相关基因PCNA的表达。结果200 cGy及以上剂量X线辐射可降低晶状体上皮细胞的增生活性,使细胞排列紊乱、固缩或脱壁;随着X线剂量的增加,各剂量组增生相关基因PCNA表达明显下降。结论200 cGy及以上剂量X射线可明显抑制晶状体上皮细胞的增生,抑制存在明显的剂量依赖关系,其作用机制可能与抑制PCNA表达有关。

Abstract:Objective To study the influence on proliferation of human lens epithelial cells induced by X-ray.Methods SRA01/04 cells after being irradiated by different dosage of X-ray(50cGy,100cGy,200cGy,300cGy,400cGy,500cGy) were gathered at 60h to observe the morphological changes,and the proliferative activity was measured by MTT assay.PCNA expression was detected by PI fluorescent staining.Results 200 cGy and over dose of X-ray could decrease the proliferation of cells,make the cells disordered arrangement,shrinkage,detachment,and PCNA expression was decreased with the increasing dosage of X-ray.Conclusion X-ray exceeding 200cGy may prohibit the proliferation of human lens epithelial cells in dose-dependent manner,which may be related to the effect of PCNA expression decreasing.
