下调血清高迁移率族蛋白1水平对急性出血坏死性胰腺炎大鼠全身毛细血管渗漏的影响

摘    要:目的 观察丙酮酸乙酯(EP)对急性出血坏死性胰腺炎(AHNP)大鼠全身毛细血管渗漏(SCL)的影响,探讨下调血清高迁移率族蛋白1(HMGB1)水平在AHNP治疗中的作用.方法 采用胰管逆行灌注人工胆汁的方法复制大鼠AHNP模型.随机分为正常对照组(Normal组,n=8)、假手术组(Sham组,n=8)、AHNP组(n=16)、EP组(n=16).EP组建模12 h开始使用EP治疗.建模24 h取材,检测与水分渗漏相关的肺系数、肺湿/干比、腹水量.使用伊文氏蓝(EBD)尾静脉注射,检测与蛋白渗漏相关的腹水、肺、胰腺、空肠、回肠、耳廓组织EBD含量;Western blot法检测血清中HMGB1的含量.结果 建模后24 h,EP组血清HMGB1浓度明显低于AHNP组(87±16)ng/ml vs.(154±22)ng/ml,P<0.05].EP组大鼠腹水量、肺系数、肺湿/干比明显低于AHNP组,均P<0.05.EP组大鼠胰腺、空肠、回肠和耳廓组织EBD含量明显低于AHNP组.结论 下调血清HMGB1水平可有效减轻AHNP大鼠SCL的程度.

Abstract:Objective To study the effect of the down-regulation of serum high mobility group box protein 1(HMGB1) expression on systemic capillary leakage(SCL) of rats with acute hemorrhagic necrotizing pancreatitis(AHNP).Methods Rat AHNP model was induced by retrograde injection of artificial bile into the pancreatic duct.Animals were randomly divided into 4 groups:normal control group(n=8),Sham group(n=8),AHNP group(n=16) and ethyl pyruvate(EP)-treated group(n=16).The rats in EP-treated group were administrated with EP(30 mg/kg,12 and 18 h post-AHNP).HMGB1 serum levels were detected by Western blotting.Evan's blue dye(EBD) was injected into caudal vein to evaluate protein leakage.The index related to water and protein leakage was detected.Results 24 h after operation,serum HMGB1 levels,ascites volume,lung coefficient,lung wet to dry ratio,EBD content in pancreas,jejunum,ileum and auricle tissue in EP group were significantly lower than in AHNP group.Conclusion Down-regulation of serum HMGB1 levels could effectively alleviate SCL in AHNP rats.
