血吸虫肝纤维化小鼠肝组织中去甲肾上腺素及受体的表达

摘    要:目的研究血吸虫肝纤维化小鼠肝组织中交感神经递质去甲肾上腺素(norepinephrine,NE)及其受体(adrenal receptor,AR)的表达。方法应用腹部敷贴尾蚴法使小鼠感染血吸虫。30只昆明种小鼠随机分成正常对照组(N组)和肝纤维化模型组(M组)。采用光镜及电镜观察肝组织病理学改变,高效液相色谱-电化学(HPLC-ECD)法检测肝组织中NE的含量,同时应用Western blot方法检测肝脏组织中AR的表达水平。结果血吸虫肝纤维化小鼠的肝组织中NE的含量明显低于正常小鼠,而AR的含量明显高于正常组,其差异具有统计学意义(P〈0.01)。结论小鼠血吸虫肝纤维化时肝组织中去甲肾上腺素表达减少,这可能是肝纤维化时交感神经促进肝纤维化发展的作用机制之一。

Abstract:Objective To investigate the expression of norepinephrine(NE) and adrenal receptor(AR) in mice with liver fibrosis induced by Schistosoma mansoni.Methods The mice were infected with schistosoma by means of pasting the cercaria to the abdomen.Thirty Kunming mice were randomly divided into control group and model group.Under the light and electron microscopy,the histopathological changes of liver were observed.The contents of NE in liver tissue were determined by high performance liquid chromatography-electrochemical detector(HPLC-ECD).The expression of AR in liver tissue was assessed by Western blot.Results The contents of NE in model group were obviously lower,but those of AR were significantly higher than that in normal group(P<0.01).Conclusion The expression of NE was decreased in liver tissue of mice with fibrosis induced by Schistosoma mansoni,Which might be one of the mechanisms by which sympathetic nerve promotes the development of liver fibrosis in mice.
