新辅助化疗对大肠癌细胞凋亡和P53表达的影响

目的:研究新辅助化疗能否引起大肠癌细胞的凋亡和对大肠癌组织P53表达的影响。方法:选取中南大学湘雅二医院老年外科2004年9月至2006年9月收治的大肠癌手术病人68例,分别采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(term inal-deoxynucleotidyl transferase med iatednick end labeling,TUNEL)和免疫组化S-P法对68例患者大肠癌组织中的肿瘤细胞的凋亡指数(Apoptotis index,AI)和P53表达进行检测。结果:新辅助化疗组肿瘤细胞的AI均数为3.56%,明显高于对照组的2.29%(P<0.01)。新辅助化疗组P53阳性表达率为22.60%,明显低于对照组的33.60%(P<0.05)。结论:新辅助化疗可以显著诱导大肠癌细胞凋亡,降低凋亡相关基因P53的表达。

Affection of Short-Term Neoadjuvant Chemotherapy on Cell Apoptosis and Expression of P53

Objective:To investigate whether neoadjuvant chemotherapy could induce tumor cell apoptosis and review its effect on expression of P53 in carcinoma of large intestine.Methods: Apoptosis index of tumor cells was assayed by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)and positive rate of P53 expression was examined by immunohistochemical S-P in 68 carcinoma of large intestine samples.Results:Tumor cell AI in neoadjuvant group(3.56%)was significantly higher than that in control group(2.29%)(P<0.01).P53 positive expression rate in neoadjuvant chemotherapy group(11/38,28.9%) was significantly lower than that in control group(17/30,56.7%)(P<0.05).Conclusion: Neoadjuvant chemotherapy could induce cell apoptosis and degrade the positive rate of P53 expression in carcinoma of large intestine.
