丙型肝炎患者T淋巴细胞亚群及调节性T细胞的变化研究

目的研究丙型肝炎患者外周血T淋巴细胞亚群和免疫调节T细胞在病程发展中的变化。方法采用流式细胞术（FCM）检测丙肝患者RNA高拷贝组、低拷贝组、RNA阴性组及健康对照组外周血CD3^＋、CD4^＋、CD8^＋及CD4^＋CD25^＋CD127^＋调节T细胞百分率，分析其在丙型肝炎病程中的变化。结果丙型肝炎患者CD4^＋T细胞降低，CD8^＋T细胞升高，CD4^＋／CD8^＋降低，患者组与对照组差异均有统计学意义（P〈0．05）；RNA阴性组和低拷贝组调节性T细胞（CD4^＋CD25^＋CD127^＋）与正常对照组明显偏高（P〈0．05），RNA高拷贝组与对照组无明显统计学差异（P〉0．05）。结论在丙型肝炎病程发展过程中T淋巴细胞亚群均发生变化且有调节性T细胞的增高，说明细胞免疫及调节机制参与了丙型肝炎的发病及发展，为临床诊疗提供了新的思路。

The study on the change of T lymphocyte subsets and Regulatory T Cells in patients with HCV infection

Objective The changes of peripheral blood T lymphocyte subsets and Regulatory T Cells in patients with HCV infection in def- ferent eourses of disease were studied. Methods Flow cytometry was used to determined the percent of CD3^＋ . CD4 ^＋、 CD8 ^＋ and CD4^＋ with HCV infection from RNA highcopy groupdowcopy group、RNA negative group and healthy control group,and the changes in the different courses were analysised. Results Significantly lower percentage of CD4^＋ T lymphocyte, and higher of CD8 ^＋ , with CD4^＋/CD8 ^＋ ratio in HCV patients than those in healthy control group（ P 〈 0.05）, and significantly higher percentage of CD25^ CD127^- T lymphocyte in patients in the low - copy group and RNA negative group than those in healthy control group（P 〈 0.05） ,but no significant differences in high -eopy group were demonstrated. Conclusion The results of the changes from T cell subsets and Regulatory T Cells suggest that the cellular immunity and the reg- ulation mechanism participate in the occurrence and development of hepatitis C, and the result will show a thinking for the clinical diagnosis and treatment to hepatitis C.
