呼吸道合胞病毒单克隆抗体用于纸上桥联酶标诊断的初步探讨

摘    要:为了呼吸道合胞病毒(RSV)能够在基层及时帮助诊断和控制其流行,作者旨在更加简化RSV的快速诊断,将RSV-McAb移在微孔滤膜上以搭桥法和RSV抗原结合,由碱性磷酸酶催化底物,在膜上显色。作者将RSV-McAb用于RSV培养物的检测,并作了异种动物血清封闭试验以及vero细胞等阴性对照,证实RSV-Mc-Ab用于纸上桥联是特异性显色,对23例患儿鼻咽部脱落细胞同时作纸上桥联,免疫荧光及APAAP 3种RSV的快速诊断的比较,获得初步满意结果。

Abstract:Respiratory syncytical virus (RSV) is one of the most important viruses in pneumonia of infan's and children. In order to diagnose and control its epidemic of the disease at the beginning in time, we wanred to simplify the rapid diagnostic method of RSV by putting the RSV-McAb with the combination with RSV-Ag on the acetic cellulose membrane. By means of the secondary antibodies as a bridge, the alkaline phosph-atase of APAAP complex catalyzes susbstrates to show the colour on the membrane. We tested RSY cultures diluted in different liter and made the oiosed experiments of different races of animal anti-RSV serum. We also made the negative comparitive experiments on vero cells, RPMI-1640, with 15% calf serum and PBS, etc. It is proved that RSV-McAb was combined with RSV-Ag specifically on cellulose membrane, rather thac other substances. We used three methods (Bridge on paper, Ilnmunofluorescence, and APAAP combination) to diagnose 23 patients. 19 of them had the same result. Only 3 patients got positive by paper method and got negative by other ones.The method of bridge on paper do not need any special instrument. The result can be dentified by naked eyes. A little reagent and specimen are used. J t is suitable for the investigation of epidemic in the rural area and the rapid diagnosis.
