川芎嗪对健康及急性心肌梗塞大鼠血流动力学影响

摘    要:目的研究川芎嗪(TMP)对正常及急性心肌梗塞(AMI)大鼠有关血流动力学的影响。方法24只SD雌性大鼠,随机分为生理盐水对照组(A组),正常川芎嗪(B组)、生理盐水对照组(C组)与AMI川芎嗪组(D组),从尾静脉注入TMP或等容量生理盐水,用RM-6200型多导生理记录仪与MF-21型电磁流量仪测取心输出量(CO),左室压(LVP),左室内压最大上升及下降速率(±LVdp/dtmax);同时观测标准Ⅱ导心电图,记录心率(HR)。结果24只大鼠无一死亡。注药前各监测指标在A、B两组及C、D两组间均无显著性差异,与A组相比,TMP可使健康大鼠出现明显的HR增快,BP下降,以及CO的增加,BP和CO改变的最大值均出现在给药即刻,5~7分钟内逐渐恢复至给药前水平,±LVdp/dtmax和时间一张力指数(TTI)为先降后升,5分钟时达最高峰;在AMI大鼠组,TMP对HR无明显改变,但出现BP下降和CO增加,对±LVdp/dtmax和TTI只有短暂的抑制,l~3分钟后恢复至用药前状态,无增强作用。结论TMP对健康及AMI大鼠血流动力学影响存在明显差异,可显著提高AMI大鼠CO,降低心肌耗氧量,为临床治疗缺血性心脏病提供了试验依据。

Abstract:Objective: To study the effeCt of tetramethyl pyraxine (TMP) on the hemodynamics of healthy tats and tats with acute myocardial infarction (AMI). Methods: A total of 24 SD tats were randoruized into control group (A), TMP treaed group (B), AMI control group (C) and AMI TMP treated group (D). TMP or equal volume of normal saline was given by injection through the caudal vein. Cardiac output (CO), blood pressure (BP), left ventricular pressure (LVP), maximum left ventricular prespure rise rate and maximum left ventricular pressure fall rate (t LVdp/dtmax) were defermined with RM - 6200 physical recorder and MF - 21 electro magnatic flow meter. ECG of standard Ⅱ and heart rate (un) were recorded at the same fime. Results: No significant difference were found befween A and B or C and D group before administration of TMP. In compare with the control group (A), TMP might increse HR and CO but decrease BP. The most marded changes of BP and CO appeared immediately agter the administration of the drug and refurned to the same leoel as premedication 5-7min later. The ± LVdp/dtmax and Time- tenson- index (TTI, rn) decreased firstly and increased later, reached the peak at 5min. In AMI group (D) no obvious changes was found in HR. but BP decreased and CO increased. The ± LVdp/dtmax and TTI were transiently inhibited and refurned to premedication level 1~3min later. Conclusion: There is marked difference of the effect of TMP on healthy and AMI rats. The may increase co and decrease the O2 consumption of myocardium.
