IL-17与Th17细胞在小鼠心脏移植 急性排斥反应中的作用

摘    要:目的:研究辅助T细胞17(T helper cell 17, Th17)细胞及其相关因子白细胞介素17(interleukin-17, IL-17)在小鼠心脏移植排斥反应中的表达及意义。方法:建立小鼠心脏移植模型,实验动物随机分为急性排斥反应组(n=12)和同系移植组(n=12),应用酶联免疫吸附实验(enzyme-linked immunosorbent assay, ELISA)分别检测2组小鼠移植术后第3天和第7天血清中IL-17的含量;应用流式细胞技术检测移植心脏中浸润淋巴细胞中Th17细胞数量,并取移植心脏经10%甲醛固定后行常规病理检查。结果:急性排斥反应组术后第3天及第7天血清中IL-17表达明显高于同系移植组(P<0.05),且急性排斥反应组术后第7天IL-17表达明显高于第3天(P<0.05);与同系移植组相比,急性排斥反应组移植心脏浸润淋巴细胞中Th17细胞比例在术后第3天及第7天明显增高(P<0.05),且术后第7天时Th17细胞比例明显高于第3天(P<0.05);病理组织学检查急性排斥反应组随着移植时间的延长,排斥反应逐渐增强。结论:Th17与心脏移植排斥反应的发生、发展密切相关,移植脏器中细胞因子IL-17的检测可望成为急性排斥反应早期诊断的预测指标。

Abstract:ObjectiveTo investigate the role of T helper cell 17(Th17) cells and their cytokines IL-17 in heart allograft rejection in mice.MethodsThe heart transplantation models were randomly divided into 2 groups: a allograft group (n=12) and an isograft group (n=12).On the post-operative day (POD) 3 and 7, serum IL-17 level was tested by enzyme-linked immunosorbent assay, and Th17 cells in heart grafts were measured by flow cytometry. The heart grafts were harvested and saved in 10% formalin, and then embedded in paraffin. ResultsCompared with the iso-graft group, the allograft group had a higher serum level of IL-17 on POD3 and POD7 (P<0.05), and the level of IL-17 was higher on POD7 than that on POD3 (P<0.05). The allograft group had more Th17 cells infiltrating in grafts on POD3 and POD7 (P<0.05) and there were more Th17 cells infiltrating on POD7 than that on POD3 (P<0.05). Histological examination showed that the prolongation of post transplantation time resulted in more rejection pathological changes. ConclusionTh17 cells may play an important role in the development of heart transplant rejection. IL-17 may serve as a predictive parameter for allograft rejection in the future.
