复方卡波姆前房注射构建大鼠慢性高眼压模型的实验研究

摘    要:目的构建适用于研究抗青光眼药物降眼压作用及保护视神经作用的大鼠青光眼模型。方法将29只SD大鼠随机分为A、B、C、D、E五组,其中A组6只,给予大鼠右眼前房注射复方卡波姆溶液5ul,B组6只给予大鼠右眼前房注射复方卡波姆溶液10ul,C组6只给予大鼠前房注射复方卡波姆溶液15ul,D组6只给予大鼠右眼烧灼3条巩膜上静脉,E组5只大鼠为空白对照组,只做前房穿刺,并对4组青光眼模型进行观察。结果A组眼压增高持续20~30d,眼压范围为(25.23±2.31)mmHg,青光眼模型成功率100%。B组眼压增高持续20~30d,眼压范围为(28.23±4.31)mmHg,青光眼模型成功率100%。C组眼压增高持续10~20d,眼压范围为(30.56±3.45)mmHg,青光眼模型成功率为100%。D组眼压增高7—10d。眼压范围为(25.54±2.45)mmHg,青光眼模型成功率为50%。E组:空白对照组眼压范围为(12.34±3.45)mmHg。结论B组大鼠在术后第1天眼压即升高,表现为升高快,持续时间长,角膜一般状况良好,副作用少,是较为理想的造模剂量。

Abstract:OBJECTIVE To build suitable model for the study of glaucoma medications effects in function and protect optic nerve in glaucoma. METHODS Twenty-nine SD rats were divided into A, B, C, D, E five groups randomly. Group A ( 6 rats) given injection of compound carbomer solution 5 ul in the fight anterior chamber, group B (6 rats) given injection of compound carbomer solution 10 ul in the right anterior chamber, group C (6 rats) given injection of compound earbomer solution 15 ul in the fight anterior chamber, group D (6 rats ) given right eye burn rats 3 veins on the sclera, 5 rats of group E as the blank control group were given anterior chamber puncture, and to look at the four groups of glaucoma model. RESULTS A group increased intraocular pressure IOP range for 20-30d, (25.23 ± 2.31 ) mmHg, the success rate of glaucoma model was 100%. B group increased intraocular pressure IOP range for 20-30d, range for 10-20d, (28.23 ±4.31 ) (30.56± 3.45) mmHg, the success rate was 100%. mmHg, the success rate was C group increased intraocular pressure IOP D group increased intraocular pressure IOP range for 7-10d, (25.54 ±2.45) mmHg, the success rate was 50%. E group: blank control group IOP range: (12.34 ± 3.45) mmHg. CONCLUSIONS Group B is the ideal model, because its intraocular pressure after first day was increased, performancing increased fast, long duration, the general condition of corneal good, less side effects.
