川芎嗪对视网膜缺血电生理及视网膜小动脉平滑肌细胞cAMP表达的影响

摘    要:目的探讨川芎嗪对正常兔眼视网膜急性缺血模型视网膜电图(ERG)最大反应、振荡电位(OPs)的影响及对兔视网膜小动脉平滑肌细胞环腺苷酸(cAMP)表达的影响。方法新西兰大白兔20只,随机分为川芎嗪组和模型组,每组10只,均予结扎右眼视神经lh制作兔视网膜急性缺血模型.造模前记录右眼ERG最大反应和OPs。川芎嗪组于视网膜缺血造模后0h、24h、48h及72h给予耳缘静脉注射川芎嗪注射液,复查ERG指标,模型组同期进行ERG检查,不予其他处理。视网膜缺血造模后72h处死动物,取右眼,免疫荧光技术观察cAMP在视网膜小动脉平滑肌细胞上的表达情况。结果视网膜缺血造模后24~72h,2组实验兔的最大反应振幅同期比较无明显差异(P〉0.05),川芎嗪组OPs振幅在缺血造模后24h、72h时明显高于模型组,差异有统计学意义(P〈0.05)。川芎嗪组缺血造模后72h时血管平滑肌细胞的cAMP表达强于模型组。结论川芎嗪对缺血视网膜的Ops振幅有改善作用,可能与促进视网膜小动脉平滑肌细胞cAMP表达,改善视网膜血循环有关。

Effect of Ligustrazine on electroretinogram and expression of cAMP in vascular smooth muscle cell of retina arteriole after retina ischemia

Abstract:OBJECTIVE To observe the effect of ligustrazine on retina ERG and Ops and expression of cAMP in vascular smooth muscle cell of retina arteriole after retina ischemia. METHODS Twenty rabbits were divided into two groups randomly: the ligustrazine group and the model group. Experimental retina ischemia-reperfusion injury was induced by ligatured optic nerve which was lasting for an hour on the right eye of all the rabbits. The ERG and Ops were recorded from ligustrazine group after Oh, 24h, 48h, 72h following retina isehemia injury. The same testing was done on the model group. Put the rabbit to death, exteriorize the right eye and observe expression of cAMP in vascular smooth muscle cell of retina arteriole by immunofluorescence. RESULTS Ligustrazine could not improve the amplitude maximum retina ERG reaction on acute ocular ischemic animal model, however it could obviously promote the recovery of Ops of retina after 24h and 72h following retinal ischemia (P〈0.05). Ligustrazine could improve expression of cAMP in vascular smooth muscle cell of retina after 72h isehemical injury. CONCLUSIONS The improved expression of cAMP in vascular smooth muscle cell of retina arteriole of Ligustrazine may be related to the Ops amplitude after retina ischemia.
