感染Vero细胞中SARS病原体的电镜观察

摘    要:目的 用透射电子显微镜(TEM)观察严重急性呼吸综合征(SARS)患者感染的Vero细胞,为SARS病原体的分离鉴定提供形态学资料。方法 标本为上海市疾病控制中心送检的SARS患者鼻咽拭子接种感染的Vero细胞.对培养液负染色标本和vero细胞超薄切片标本进行TEM观察。结果 在负染色标本和超薄切片标本中均可观察到病毒.病毒颗粒为圆形或椭圆形,外周有冠状排列的纤突,病毒颗粒直径约80~160nm,纤突长度约20~40nm、感染细胞的病毒颗粒分布在细胞质中,主要在内质网和空泡内,可见群集的大小较一致的病毒颗粒。结论 本例电镜所见病毒颗粒大小、形态与文献报道的SARs冠状病毒(SARS—CoV)相符。

Abstract:Objective To utilize electron microscopic study of infected cells to identify the pathogenic agent of severe acute symdrome (SARS). Methods Vero cells were collected with nasopharygeal swabs from SARS patients in Shanghai. The supernatant and infected cells were prepared for electron microscopic study. Results Nagative-stain and ultrathin-seetion revealed the characteristic coronavirus particles. The virus particles of infected cells were in the endoplasmic reticulum and vesicules. They were mainly spherical or oval in shape, 80-160 nm in diameter, with club-shaped surface projections 20-40nm. Conclusion This study indicates a novel coronavirus is the causative agent of the SARS patient.
