摘    要:目的:采用RNA干扰(RNA interference,RNA i)技术,利用构建的稳定转染CD147 shRNA表达质粒载体的HO-8910pm/pGE-1 CD147shRNA细胞株观察卵巢癌细胞系HO-8910pm CD147基因的沉默作用及生物学效应。方法:用免疫细胞化学法、激光共聚焦检测HO-8910pm细胞CD147的表达;MTT法绘制细胞生长曲线,观察裸鼠卵巢癌皮下移植瘤的成瘤性。结果:pGE-1CD147shRNA转染人卵巢癌细胞系后,CD147的表达受到高效且特异的抑制,细胞侵袭力减弱,荷瘤裸鼠成瘤力下降,抑瘤率达58.8%(P<0.01)。结论:RNA i沉默CD147基因可影响卵巢癌的侵袭和成瘤,有可能成为治疗卵巢癌的新途径。

Abstract:Objective:To study the effect of CD147 gene silence by RNAi in ovarian cancer cell line HO-8910pm on biological behavior.Methods:Immunohistochemical technique and confocal laser scanning microscope(CLSM) were carried out to assess the expression of CD147 in HO-8910pm cell line.MTT assay was used to observe the effect on proliferation ability;in vitro invasion assay was performed to assess changes of tumor invasion ability.Cells were transplanted subcutaneously in nude mice to observe the tumorigenesis.Results:In HO-8910pm/pGE-1 CD147shRNA cell,the expression of CD147 was inhibited specifically and efficiently;no statistical difference of their proliferation abilities was observed but their invasion abilities and tumorigenesis in nude mice decreased significantly.Tumorigenicity inhibition rate reached 58.8%(P<0.01) in experimental group.Conclusion:Silencing of CD147 by RNAi can inhibit invasion and tumorigenesis of ovarian cancer and may become a new approach of ovarian cancer therapy.
