内皮素及其受体在兔不全梗阻性膀胱的实验研究

摘    要:目的:内皮素及其受体在兔不全梗阻性膀胱中的改变和意义.方法运用放射免疫分析技术对10只成年雄性新西兰白兔不全梗阻性膀胱(兔不全梗阻性膀胱[1,2]为实验组分梗阻3周组及梗阻6周组)及10只同龄雄性新西兰白兔无梗阻性膀胱(为对照组,分为术后3周组和6周组)的兔血桨、尿液及膀胱平滑肌组织中内皮素-1及其受体进行研究.结果实验组较对照组血浆与尿液中内皮素-1含量均明显增高(血浆3周组与血浆6周组均为P<O.05;尿液3周组与6周组均为P<0.001);实验组较对照组膀胱体部平滑肌组织内皮素及其受体含量明显升高(P<0.05),而膀胱颈部平滑肌组织内皮素-1及其受体含量无显著性差异(P>0.2);实验组膀胱重量明显增加(P<0.001).结论内皮素-1具有收缩平滑肌和促平滑肌、纤维结缔组织增生(有丝分裂)作用,内皮素及其受体的上调在兔不全梗阻性膀胱中为其膀胱壁结构改变和功能紊乱的重要因素.血浆、尿液中内皮素在兔不全梗阻性膀胱中含量的上调与兔不全梗阻性膀胱壁结构的改变与功能紊乱的病理过程具有一定的内在联系,并可作为膀胱出口梗阻诊断的重要参数.

Abstract:Objective: To investigate the endothelin-1 and its receptor in the rabbit model of partial bladder outlet obstruction. Methods: We studied changes endothelin-1(in plasma, urine and bladder smooth muscle) and its receptor (in bladder smooth muscle) in a rabbit model of partial bladder outlet obstruction with radioimmunoassay and radio ligand and Binding Assay of Receptors. Partial bladder outlet obstruction was induced in adult male New Zealand White rabbits (ten rabbits as experiment group, time of obstruction for three weeks and six weeks). Sham operated age-matched rabbits acted as control (ten rabbits as control group, time of postoperation for three weeks and six weeks). Results: Endothelin-1 in blood plasma and urine were more increase in the time of obstruction for the three weeks experiment group than in the time of postoperation for the three weeks control group, endothelin-1 in plasma and urine were more increase in the time of obstruction for the six weeks experiment group than in the time of postoperation for the six weeks control group. Endothelin-1 in bladder smooth muscle body of the experiment group more increase than the control group. No difference in endothelin-1 in bladder neck smooth muscle has been observed the experiment group and the control group at six weeks. Weight of bladder was heavier in the experiment group than the control group. Compared with the control group, the concentration of endothelin receptor of experiment group is significantly increased. The KD values of ET-1 receptor of SMC of the body of bladder have no difference between the experiment group and the control group. The concentrations of ET-1 receptor of SMC of the neck of bladder have no difference between the experiment group and the control groups. The KD values values of ET-1 receptor of SMC of the neck of bladder have no difference between experiment group and control group. Conclusions: The up-regulation of endothelin-1 in blood plasma and urine are associated with altered bladder structure and dysfunction of rabbit partial bladder outlet obstruction and they are diagnosis methods for partial bladder outlet obstruction. The increased endothelin-1 and its receptor in smooth muscle of bladder body is associated with altered structure and dysfunction of rabbit partial bladder outlet obstruction.
