摘    要:目的建立测定人血浆中烟酸浓度的方法。方法36名健康男性志愿者口服烟酸缓释片,用反相离子对高液相色谱法测定血浆中烟酸浓度。结果烟酸血浆浓度在0.05~2.00和2.00~32.00mg.L-1范围内,有良好的线性关系, 绝对回收率为 80.5%~95.5%,日内、日间RSD小于5%,最低检测浓度为0.05mg.L-1。结论本法具有灵敏、准确、回收率高的优点,适用于烟酸人体药代动力学的研究。

Abstract:Objective To establish a reverse phase ion pair high performance liquidchromatography method for the determination and pharmacokinetic study ofnicotinic acid sustained-release tablet in Chinese healthy volunteers. Methods Theplasma samples from the 36 volunteers after taking 1 500mg nicotinic acid sustainedrelease tablets were extracted with acetone and chloroform. Human blood plasmawas precipitated with acetone, after mixing and centrifugation, the supernatant waswashed with chloroform, after mixing and centrifugation, the 0.5mL supernatantwas blowed by N2. The residue was dissolved by 0.2mL mobile phase, a 10mL ofthe solution was injected into an ODS2 column monitored with UV detector at265 nm. Results A good linearity was obtained in the range of 0.05~2.00 and2.00~32.00mg.L-1. The absolute recovery rates were between 80.5%~95.5%.The RSD of within-day and between-day were less than 5% (n=5). The minimaldetectable concentration in plasma was 0.05 mg.L-1. Conclusion The establishedmethod is sensitive, accurate and the rate of recovery is high. It is suitable for thepharmacokinetic study of nicotinic acid.
