GDNF基因诱导骨髓间充质干细胞向神经细胞分化的实验研究

摘    要:目的 观察在体外培养的经GDNF基因诱导的骨髓间充质干细胞(BMSCs)向神经细胞分化的表达.方法 体外培养大鼠骨髓间充质干细胞,选取第3代细胞,应用GDNF基因诱导BMSCs分化两周,观察诱导后细胞的形态学变化,免疫荧光方法验证及鉴定神经元特异性烯醇化酶(NSE)及髓磷脂酸性蛋白2(MAP-2)的表达.结果 镜下观察细胞形态学以及特异性荧光可见BMSCs向神经细胞分化明显.结论 GDNF基因转染的BMSCs在体外培养情况下可转化为神经细胞,并表达特定神经细胞标志蛋白.

The experimental research of cells differentiation of bone marrow mesenchymal stem cells induced by GDNF gene to nerve cells

Abstract:Objective To observe the expression of cell differentiation of bone marrow mesenchymal stem cells(BM-SCs) induced by GDNF gene to nerve cells in vitro culture.Methods Culture rat bone marrow mesenchymal stem cells in vitro,select the third generation of cells,and apply GDNF gene to induced BMSCs to differentiation for two weeks.Observe the cell morphology after induction,Identify the expression of neuron specific enolization enzyme (NSE) and myelin acidic protein 2 (MAP-2) with immunofluorescence method.Results The microscopic observation about cell morphology and specific fluorescence visible can be seen obviously that BMSCs divide to nerve cells.Conclusion BMSCs transfected by GDNF gene in vitro culture condition could be converted into a nerve cell,and some nerve cell could be expressed by special marker protein.
