葛根素抑制 D-半乳糖致大鼠蛋白糖基化的实验研究

摘    要:目的:采用D-半乳糖诱导的糖基化模型大鼠,研究葛根素(Radix Puerariae,RP)对蛋白糖基化反应的抑制作用。方法:D半-乳糖(150 mg/kg,ip)给药8周,诱导糖基化模型大鼠,并于第2周开始给予葛根素高(300 mg/kg)、中(150 mg/kg)、低(75 mg/kg)剂量处理6周。测定红细胞醛糖还原酶活性、糖化血红蛋白含量和血清果糖胺、晚期糖基化终末产物含量及胰岛素水平、SOD活性和MDA含量。结果:葛根素高、中剂量可明显降低D-半乳糖引起的模型大鼠血糖升高(P<0.01)及红细胞醛糖还原酶活性,并显著抑制糖基化产物的形成(P<0.01);同时,还可改善胰岛素敏感性,增加超氧化物歧化酶活性(P<0.01),而葛根素高剂量组可显著降低血清脂质过氧化物含量(P<0.01)。结论:葛根素对D-半乳糖诱导的大鼠糖基化反应具有明显的抑制作用。

Abstract:Objective: To observe the effects of Radix Puerariae on protein glycation in model rats induced by D-galactose.Methods: The model rats of protein glycatin were induced by intraperitoneal administration of D-galactose(150 mg/kg) for 8 weeks,and all rats were treated by Radix Puerariae(High dose 300 mg/kg,Middle dose 150 mg/kg,Low dose 75 mg/kg) for 6 weeks.The activity of aldose reductase in red blood cells,the content of fructosamine in serum,the amount of glycohaemoglobin and advanced glycation end-products,the level of insulin in serum,the activity of superoxide dismutase and the amount of maleic dialdehyde were measured. Results: High dose and middle dose of Radix Puerariae could decrease the level of blood glucose and the activity of aldose reductase in red blood cells,inhibit the formation of glycation products significantly in model rats induced by D-galactose(P<0.01),increase insulin sensitivity and activity of superoxide dismutase(P<0.01) and decrease the amount of maleic dialdehyde(P<0.01).Conclusion: Puerariae can significantly inhibit glycation reaction in rats induced by D-galactose.
