人胃癌裸鼠移植瘤DNA含量异质性的研究

摘    要:两株人胃癌细胞MKN-28和MKN-45,分别经皮下、腹腔、胃壁和尾静脉接种在无胸腺裸小鼠体内。对不同接种部位形成的胃癌移植瘤,用流式细胞仪分析了瘤细胞的DNA含量。结果证实,胃癌移植瘤细胞的DNA含量存在异质性。由此提示,在人胃癌裸鼠移植瘤的发展过程中,伴有瘤细胞DNA含量的异质性改变。这种变化也受接种部位器官环境的影响。

Abstract:The DNA content heterogeneity of human gastric cancer xenograft in ath-ymic nude mice was determined by using flowcytometer (FCM) analysis. All of the tumors from different sites of inoculation were composed of at least two subpopulations with different DNA content. Most of them had a combination of two DNAploidy patterns, one similar to that of inoculated tumor cells, and the other, a newly emerged ploidy pttern different from the former. The data indicated that threre is a heterogenenous change of DNA content of inoculated human gastric cancer cells during the prcess of tumor progression, and the DNA content heterogeneity of human gastric cancer xenograft is not only deter mined by genetic instability of tumor cells, but also influenced by the environment in athymic nude mice.
