MCP-1在子宫内膜异位症在位内膜的表达

摘    要:目的探讨MCP-1在子宫内膜异位症(EMT)患者的在位子宫内膜中的表达水平及其在内异症发病中的作用。方法 2008年5月~2009年5月在南通大学附属医院妇产科以正常子宫内膜为对照,采用免疫荧光方法与Western blot分析内异症患者在位内膜的表达水平,并比较其差异。结果 1)免疫荧光检测结果:MCP-1在内膜腺体和间质细胞中均有表达,主要定位于内膜腺体的腺泡浆。正常子宫内膜分泌期MCP-1的表达(16.1826±3.0363)与其增生期(14.7431±2.0946)相比,组间比较无显著性差异(P>0.05);内异症在位内膜分泌期MCP-1的表达(51.0695±5.3293)高于其增生期(30.0012±4.3439),组间比较存在显著性差异(P<0.01);内异症在位内膜MCP-1的表达高于正常子宫内膜(P<0.01)。2)Western blot结果显示:正常子宫内膜分泌期MCP-1的表达(0.36788±0.02522)与其增生期(0.35678±0.02984)相比,组间比较无显著性差异(P>0.05);内异症在位内膜分泌期MCP-1的表达(1.06894±0.10542)高于其增生期(0.53686±0.02303),组间比较存在显著性差异(P<0.01);内异症在位内膜MCP-1的表达高于正常子宫内膜(P<0.05)。结论 MCP-1可能在子宫内膜异位症的病变中起重要作用。

Expression of Monocyte Chemotactic Protein-1 in EMT Eutopic Endometrium

Abstract:Objective To investigate the expression of MCP-1 in EMT eutopic endometrium and the role of MCP-1 in the onset of EMT. Methods inmunohistochemistry and western blot were used to detect the proein expression of MCP-1 in EMT eutopic endometrium,compared with the endometrium of nomal fertile women as control in the affiliated hospital of Nantong University from May,2008 to May,2009. Results Immunofluorescence: MCP-1 had expression in both gland and stroma of all tissue sections,and the expression mainly located in gland.The expression of MCP-1 showed no difference between the proliferative phase(14.7431±2.0946) and secretory phase(16.1826±3.0363)in normal endometrium(P>0.05).That expression in secretory phase(51.0695±5.3293) was higher than that in proliferative phase(30.0012±4.3439) in EMT eutopic endometrium(P<0.01).Compared with the normal endometrium,the expression level of MCP-1 in EMT eutopic endometrium was increased(P<0.01)in both secretory phase and proliferative phase.Western blot: The expression of MCP-1 showed no difference between the proliferative phase(0.35678±0.02984) and secretory phase(0.36788±0.02522)in normal endometrium(P>0.05).The significant difference existed between secretory phase(1.06894±0.10542)and proliferative phase(0.53686±0.02303) in EMT eutopic endometrium(P<0.01).Compared with the normal endometrium,the expression level of MCP-1 in EMT eutopic endometrium was increased(P<0.05) in both secretory phase and proliferative phase. Conclusions MCP-1 may play an important role in the pathogenesis of endometriosis.
