阿莫西林咀嚼片在健康人体内的药代动力学研究

目的建立测定人血浆中阿莫西林浓度的高效液相色谱法。方法色谱柱:KromasilC18(250mm×4.6mm,5μm);流动相:乙腈-5mmol/L醋酸铵(1∶9);检测波长:225nm;阿司匹林作为内标,用HPLC法测定人血浆中阿莫西林浓度。结果阿莫西林线性范围为0.40～18.0μg/ml,回归方程Y=0.0048,C-0.0149(r=0.9996);回收率>80%,日内和日间RSD均<10%。结论本方法简便、快速、准确,适用于阿莫西林咀嚼片药代动力学研究及血药浓度监测。

Study on the pharmacokinetics of Amoxicillin chewable tablets in healthy volunteers

Objective To establish a high performance liquid chromatographic method for determining the concentration of Amoxicillin in healthy volunteers.Methods The assay was conducted on the Kromasil C18(250 mm×4.6 mm,5 μm) column with acetonitrile-5 mol/L ammonium acetate(1:9)as the mobile phase.The detection wave length was set at 225 m,and aspirin was taken as the internal standard.Results The linearity range was 0.40~18.0 μg/ml with a regressive equation of Y=0.0048 C-0.0149(r=0.9996).The recovery was above 80%,with both intra-day RSD and inter-day RSD below 10%.Conclusions The method is shown to be convenient,rapid,accurate,and suitable for the pharmacokinetic study of Amoxicillin chewable tablets and monitoring its concentration of clinical application.
