Bcl-2/Bax基因在非小细胞肺癌中的异常表达及其与吸烟的关系

摘    要:目的探讨凋亡相关基因Bcl-2/Bax在非小细胞肺癌(NSCLC)中的表达及其与吸烟的关系。方法应用免疫组化S—P法对33例NSCLC及正常对照肺组织中的Bcl-2和Bax表达水平进行检测,并比较其与吸烟及临床病理特征的关系。结果Bcl-2和Pax蛋白在NSCLC中均有异常表达,Bcl-2为高表达,Bax为低表达;二者的表达均与吸烟有关,Bcl-2表达吸烟组高于非吸烟组,Bax表达吸烟组低于非吸烟组。Bcl-2和Bax在非小细胞肺癌中的表达与肺癌的组织学类型及淋巴结转移情况有关,Bcl-2表达鳞癌高于腺癌,淋巴结转移组高于无淋巴结转移组;Bax表达则鳞癌低于腺癌,淋巴结转移组低于无淋巴结转移组,二者呈负相关。结论凋亡相关基因Bcl-2/Bax在NSCLC中均有异常表达,且与肺癌的组织学类型及淋巴结转移有关,说明凋亡作用失衡在NSCLC发生发展中起重要作用。Bcl-2/Bax表达与吸烟有关,提示吸烟可影响机体的细胞凋亡,参与肺癌的发生。

Abstract:Objective: To study the relationship beteen expressions of apoptosis associated genes Bcl-2/Bax in non small cell lung cancer (NSCLC) and smoking. Methods: Immunohistochemical S-P technique was used to detect Bcl-2/Bax protein expression in 33 cases NSCLC tissues and normal control lung tissues, then study the relationship between their expression and smoking, pathological characteristics. Results: Both Bcl-2/Bax showed abnormal expression in NSCLC tissue, Bcl-2 showed high expression and Bax showed low expression. the expression of the two genes were associated with smoking, Bcl-2 showed high expression and Bax low expression in smoking group. These two genes had negative correlation with each other. Bcl-2 expression in squamous cell carcinoma was higher than that in adenocarcinoma. The group with lymph nodes metastasis was higher than that without lymph nodes metastasis. Bax protein expression in squamous cell carcinoma was lower than that in adenocarcinoma. The group with lymph nodes metastasis was lower than that without lymph nodes metastasis. Conclusions: Apoptosis associated gene Bcl-2/Bax shows abnormal expressions in NSCLC, and the expressions are associated with pathological characteristics and lymph node metastasis. The expression of Bcl-2/Bax in NSCLC is associated with smoking, and probably smoking can affect apoptosis and then play a role in the process of carcinogenesis.
