姜黄素对肝癌细胞分化相关代谢指标的影响

摘    要:【目的】研究姜黄素对肝癌细胞BEL-7402诱导细胞分化的机制。【方法】用酶促反应试剂盒检测细胞浆中γ-谷氨酰转肽酶(γ-GT)活性,免疫组化染色检7714细胞浆中甲胎蛋白(AFP)合成情况,放免法检测细胞AFP和白蛋白(ALB)分泌量。观察不同浓度姜黄素对以上指标的影响。【结果】①各实验组γ-GT活性在培养过程中逐渐下降,自d2开始显著下降,d3~5缓慢下降,且较同期培养的对照组显著性降低(P〈0.01)。②细胞免疫组化结果显示,对照组细胞AFP表达较强,着色较深,呈棕黄色,主要成团浓集分布于胞浆中。实验组AFP在胞浆内均匀分布,呈黄色,着色较淡。随姜黄素浓度的增加而阳性程度减弱。③各实验组AFP分泌量均于d2显著下降,d5降至最低值。实验组细胞AFP分泌量较同天培养的对照组细胞显著降低(P〈0.01)。④对照组ALB分泌量自d2开始明显升高,自d5开始有小幅度下降,实验组细胞ALB分泌量较同天培养的对照组细胞显著性升高(P〈0.01)。【结论】姜黄素可影响肝癌细胞BEL-7402代谢指标,诱导其在体外向正常肝细胞分化。

Changes of Metabolism Indicators Related to Cell Differentiation of Hepatocarcinoma Induced by Curcumin

Abstract:[Objective]To investigate the mechanism of curcumin for inducing cell differentiation of human hepatocar- cinoma cell line BEL-7402. [Methods] Enzymatic reaction kit was used to measure the activity of γ-glutamyl transpeptid- ase(γ-GT) in cytoplasm. Immunohistochemical staining was used to detect the synthesis of alpha-fetoprotein(AFP) in cytoplasm. Radioimmunoassay was used to examine the secretory volume of AFP and albumin(ALB). The effect of cureu- min on above indicators was observed. [Results]After human hepatocarcinoma cell lines BEL-7402 was treated with vari- ous concentration of curcumin, the activity of γ-GT in all experimental groups during the culture gradually decreased, aad significantly decreased at 2d and slowly decreased at 3-5d. Compared with control group, the activity of γ-GT in all ex- perimental groups significantly decreased( P 〈0.01). Immunhistochemistry result showed that the expression of AFP in the cells of control group increased, and was buffy-yellow with deep color and mainly distributed in cytoplasm. In experi- mental groups, AFP was evenly distributed in cytoplasm and yellow with light color. With the increasing of the drug dose, the positive expression of AFP decreased. The secretory volume of AFP in hepatocarcinoma cell of experimental groups significantly decreased at 2d and dropped to the lowest level at 5d. Compared with control group, the secretory volume of AFP in experimental groups at the same day significantly decreased( P 〈0.01). The secretory volume of ALB in control group obviously increased at 2d and slightly decreased at 5d. Compared with control group, the secretory vol- ume of ALB in experimental groups at the same day significantly increased( P 〈0.01).
