健脾活血补肾汤对CIA大鼠血清IL-17及OPG/RANKL表达的影响

摘    要:目的通过观察健脾活血补肾汤对关节炎(collageninduced arthritis,CIA)模型大鼠血清中炎性细胞因子白细胞介素-17(interleukin-17,IL-17)、骨保护蛋白(osteoprotegerin,OPG)及核因子KB受体活化因子配体(receptor activator for nuclearfactor—KBligand,RANKL)表达水平的影响,探索健脾活血补肾汤保护强直性脊柱炎关节骨质破坏潜在的分子药理机制。方法利用弗氏完全佐剂和牛Ⅱ型胶原建立大鼠关节炎(CIA)模型,自造模成功后第3天起分别按健脾活血补肾汤高、中、低剂量给药组及生理盐水给予大鼠灌胃处理,每天1次,连续灌胃30d,处死大鼠,心脏采血取血清,用ELISA夹心法检测大鼠血清中IL~17、0PG及RANKL表达水平。结果健脾活血补肾汤高、中、低各剂量组CIA大鼠血清IL-17及RANKL水平均较模型组明显降低,差异有统计学意义(P〈0.05);而OPG水平较模型组显著升高,差异有统计学意义(P〈0.05)。结论健脾活血补肾汤可能通过降低CIA模型大鼠血清中IL-17及RANKL表达水平,升高OPG表达水平,而起到延缓关节炎性损伤所致的骨质破坏的作用。

Effects of Intensifying Kidney and Spleen and Activating Blood Circulation Decoction on the Express of IL-17 and OPG/RANKL in Serum of CIA Rat

Abstract:Objective To explore the effects of intensifying kidney and spleen and activating blood circulation decoction on the express of IL-17 and OPG/RANKL in serum of CIA rat. Methods The CIA rat model was induced by intradermal of CFA and type Ⅱ collagen. A total of 60 Wistar rats were divided into 5 groups randomly, three treatment groups which were given high, medium and low concentration of intensifying kidney and spleen and activating blood circulation decoction respectively, control group which was given normal saline and CIA mode group. After being lavaged for 30 days,the level of IL-17,OPG and RANKL in serum was determined by ELISA kits. Results Compared with the model group, the level of IL 17 and RANKL in rat serum of three treatment groups decreased significantly(P〈0.05),whereas the serum level of OPG in three treatment groups increased markedly(P〈0.05).
