吴茱萸水溶性成分指纹图谱研究

摘    要: 目的建立吴茱萸水溶性成分的HPLC指纹图谱,为鉴定吴茱萸药材提供依据。方法色谱柱:YWG-C18(250mm×4.6mm,10μm),流动相:乙腈-0.4%磷酸梯度洗脱,流速:0~8min为1.0mL·min-1,8-20min为0.5mL·min-1,20-35min为1.0mL·min-1,检测波长327nm,柱温30℃,进样量20μL。结果测定的10批吴茱萸样品有15个共有峰,共有峰相对保留时间的RSD为0.24%~1.3%,相对峰面积的RSD为11.8%~59.3%。10批样品之间的重叠率均在0.80以上,共有峰占总峰面积均约在90%。结论本实验首次建立了中药吴茱萸中水溶性成分的HPLC指纹图谱,为吴茱萸药材的真伪鉴别提供了科学依据。

Abstract:OBJECTIVE To establish a HPLC fingerprint of water soluble component in Evodia rutaecarpa (Juss. )Benth for identifying and evaluating its quality. METHODS Water extract of Evodia rutaecarpa(Juss.) Benth was separated on a YWG-C18 column (250mm×4.6 mm,10μm).The mobile phase consisted of acetonitrile-0.4% phosphoric acid with gradient elution. The flow rates were 1.0 mL·min-1(0-8min),0.5 mL·min-1(8-20 min) and 1.0 mL·min-1 (20-35 min).The wavelength of UV detection was 327 nm.The column temperature was at 30℃ and 20 μL was injected. RESULTS There were 15 common peaks in the fingerprint of 10 samples. The RSDs of the relative retention time of the common peaks was 0.24%-1.3%. The RSD of the relative peak area was 11.8% -59.3% .The overlap rates of the 10 samples were more than 0.80.The percent of the common peaks area in total peaks area were about 90%.CONCLUSION The fingerprint of water soluble component in Evodia rutaecarpa (Juss.) Benth was firstly established. It will be used to identify and evaluate the quality of Evodia rutaecarpa(Juss.)Benth.
