Rs2880412位点多态性与肥胖易感性的病例对照研究

摘    要:目的 探讨PYY基因rs2880412位点多态性与汉族肥胖人群易感性的关系。方法 抽取华中地区汉族人群成年肥胖患者410例,按照年龄(±3)、同性别匹配选取410例非肥胖人群作为对照,进行体格检查及血液生化指标检测,PCR-RFLP法进行基因多态性检测。结果 检出rs2880412位点等位基因A和C,且在对照组和肥胖组中的分布差异有统计学意义(P<0.05),等位基因A作为参照,等位基因C是肥胖的易感性等位基因(OR=1.29,95%CI:1.06~1.56);检出rs2880412位点基因型AA、AC和CC,其中基因型CC和AA在对照组和肥胖组中的频率分布差异具有统计学意义(P均<0.05);基因型CC在肥胖组和对照组中的频率分布差异具有统计学意义(P<0.05),调整混杂因素年龄、性别后,CC、AC、AC+CC基因型的OR(95%CI)分别为1.74(1.16~2.62)、1.19 (0.86~1.64)和1.32(0.97~1.79)。不同基因型生化指标分析结果显示,AA基因型的TG和LDL-C指标均值分布差异有统计学意义(P均<0.05)。结论 PYY基因rs2880412位点C可能与汉族人群肥胖发生存在关联,AA可能与肥胖组人群TG升高和LDL-C降低存在关联。

Abstract:Objective To investigate the relationship between polymorphism of PYY gene rs2880412 and susceptibility to obesity in Han Chinese. [WTHZ]Methods [WTBZ]410 adult obese patients of Han nationality in central China were selected, and 410 non-obese patients were selected according to age (±3) and gender matching as controls. Physical examination and blood biochemical indexes were performed on the subjects, and gene polymorphism was detected by PCR-RFLP method. [WTHZ]Results [WTBZ]Alleles A and C were detected at rs2880412, and the difference in distribution between the control group and the obesity group was statistically significant (P<0.05). Compared with allele A, allele C was the susceptibility allele to obesity (OR=1.29, 95%CI: 1.06~1.56). Three genotypes AA, AC and CC were detected at rs2880412 locus, and the differences of genotypes CC and AA in the control group and the obesity group were statistically significant (all P<0.05). With genotype AA as the reference, the distribution difference of genotype CC was statistically significant (P<0.05). After adjusting for age and gender, the OR(95%CI) of CC, AC and AC+CC genotypes were 1.74 (1.16~2.62), 1.19 (0.86~1.64) and 1.32 (0.97~1.79), respectively. The results of biochemical indexes analysis of different genotypes showed that the mean distribution differences of TG and LDL-C indexes of AA genotype were statistically significant (all P<0.05). [WTHZ]Conclusion [WTBZ]PYY gene rs2880412 locus C may be associated with obesity in human population, while AA may be associated with TG increase and LDL-C decrease in obese group.
