水飞蓟和葛根复合物对大鼠酒精性肝纤维化的影响研究

摘    要:目的 建立酒精性肝纤维化大鼠模型,探讨水飞蓟和葛根复合物对大鼠酒精性肝纤维化的影响。方法 通过给予改良的L/D乙醇液体饲料和腹腔注射CCl4橄榄油溶液的方法建立大鼠酒精性肝纤维化模型。确定造模成功后,将模型组动物随机分为低、中、高剂量组、模型对照组和阳性对照组,同时设置空白对照组。各组灌胃给予相应剂量的受试物持续30 d,空白对照组喂饲基础饲料,其余组喂饲改良的L/D乙醇液体饲料。实验结束后检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转肽酶(GGT)和甘油三脂(TG)含量,取部分肝脏做病理切片,HE和Masson染色后镜检,进行评分。统计分析采用 SPSS 20软件,多组间均数比较用单因素方差分析,组间比较采用Dunnett - t检验。结果 模型组ALT含量、肝脏纤维化病理评分较空白组均升高,且差异有统计学意义(F = 10.022,P<0.05;F = 6.297,P<0.05);水飞蓟葛根复合物各剂量组肝脏纤维化病理评分较模型组降低,且差异有统计学意义(F = 6.297,P<0.05)。结论 模型建立成功,水飞蓟葛根复合物可以改善酒精性肝纤维化。

Abstract:The study was designed to establish a rat model of alcoholic liver fibrosis and to investigate the effects of Pueraria lobate combined with Silybum marianum on improving alcoholic liver fibrosis. Methods The model groups were built by giving improved L/D ethanol liquid feed and intraperitoneally injected the CCl4 olive oil solution. After identification of liver fibrosis models, the animals of model group were randomly divided into three dose groups of low, medium and high level of P.lobate combined with S.marianum, model control group and positive control group. The blank group was also set.All groups were treated by intragastric administration for 30 days. The blank control group was given basic feed, all other groups were continuously given improved L/D ethanol liquid feed until the end of the experiment. The serum level of Alanine aminotransferase(ALT), Aspartate aminotransferase(AST), Alkaline phosphatase(ALP),-glutamyl transpeptidase(GGT) and Triglyceride(TG) were all examined after the experiment, and the liver was taken for pathological examination and scored with H&E and Masson staining. Statistical analysis was performed by SPSS 20. The comparison between the mean of multiple groups was analyzed by one-way analysis of variance, and the comparison between groups was performed by Dunnett-t.Results The ALT content and the pathological score of liver fibrosis of the model group were higher than those of the blank group(F=10.022, P<0.05; F=6.297, P<0.05). The pathological score of liver fibrosis in each of the dose groups was lower than that of the model group(F=6.297, P<0.05). Conclusion The animal models are successfully built. The combination of Pueraria lobate and Silybum marianum has the effect of improving the fibrosis of liver tissue.
