CHD1L、ECRG4蛋白在早期宫颈鳞癌中的表达及临床意义

摘    要:目的:探讨CHD1L、ECRG4蛋白表达与早期宫颈鳞癌恶性生物学行为的关系。方法:免疫组化SP法检测48例宫颈鳞癌、40例宫颈鳞状上皮内病变(SIL)及20例正常宫颈组织中CHD1L、ECRG4蛋白的表达情况及定位分布,并结合宫颈鳞癌患者临床病理资料进行分析。结果:CHD1L蛋白在正常宫颈组织、SIL、宫颈鳞癌组织中呈递增表达,而ECRG4蛋白则呈递减表达,差异均有统计学意义(P<0.05)。早期宫颈鳞癌组织中,CHD1L蛋白阳性率与肿瘤FIGO分期及淋巴结转移呈正相关(P<0.05),而与患者年龄、肿瘤分化程度无关(P>0.05);ECRG4蛋白阳性率仅与肿瘤分化程度呈负相关(P<0.05),而与患者年龄、FIGO分期及淋巴结转移无相关性(P>0.05)。Spearman等级相关分析表明,宫颈鳞癌中CHD1L与ECRG4蛋白表达水平呈负相关(r=-0.402,P<0.05)。结论:CHD1L与ECRG4负相互作用,共同参与了宫颈鳞癌的发生与恶性化过程,有望成为监测宫颈鳞癌进展的潜在分子标记物。

Abstract:Objective:To explore the association of CHD1 L and ECRG4 expressions with the malignant biological behaviors in early stage cervical squamous cell carcinoma.Methods:Immunohistochemical method(SP) was used to detect the expression and localization of CHD1 L and ECRG4 proteins in 48 cases of cervical squamous cell carcinoma,40 cases of SIL and 20 cases of normal cervical tissue.The expressions of CHD1 L and ECRG4 in cervical squamous cell carcinoma were analyzed with clinicopathologic data.Results:The expression of CHD1 L protein was increased gradualy in normal cervical tissue,SIL and cervical squamous carcinoma,while ECRG4 was contrary expressed(P<0.05).In early stage cervical squamous cell carcinoma,the positive rate of CHD1 L was related to FIGO stage,lymph node metastasis(P<0.05),but had no statistical relationship with the age of patients and degree of tumor differentiation(P>0.05);while the positive rate of ECRG4 was related to degree of tumor differentiation(P<0.05),but had no related with the age of patients,FIGO stage and lymph node metastasis(P>0.05).In early cervical squamous cell carcinoma,the expressions of CHD1 L and ECRG4 were negatively correlated(r=-0.402,P<0.05).Conclusion:The expressions of CHD1 L and ECRG4 were negatively correlated,they worked together to affect the malignant biological behavior of early stage cervical squamous cell carcinoma and would become potential molecular markers in predicting disease progression.
