脂质体槲皮素对小鼠结肠腺异位实体瘤的影响研究

摘    要:目的考察脂质体槲皮素对人结肠癌腺细胞株HT29 BALB/c小鼠异位实体瘤的作用。方法建立HT29结肠腺癌BALB/c小鼠肿瘤模型,将48只成模小鼠随机分成模型组、脂质体槲皮素组、槲皮素组、5-氟尿嘧啶组,每组12只。接种后4 d,模型组小鼠腹腔注射生理盐水0.3 mL,脂质体槲皮素组腹腔注射脂质体槲皮素50 mg/kg,槲皮素组腹腔注射槲皮素50 mg/kg,5-氟尿嘧啶组腹腔注射5-氟尿嘧啶15 mg/kg,每隔3 d注射1次,共注射8次。干预过程中记录各组小鼠肿瘤体积和体质量;末次注射后3 d处死各组小鼠,剥离肿瘤,称质量,测肿瘤体积,计算瘤体积抑制率和瘤质量抑制率,HE染色观察肿瘤病理表现。结果各组小鼠均无死亡,脂质体槲皮素组、槲皮素组的肿瘤体积均明显小于模型组(P均<0.05),但各组小鼠体质量比较差异无统计学意义(P均>0.05);脂质体槲皮素组、槲皮素组、5-氟尿嘧啶组的瘤体积抑制率分别为25.43%,22.27%,26.06%,瘤质量抑制率分别为15.47%,13.37%,16.66%,脂质体槲皮素组和5-氟尿嘧啶组均明显高于槲皮素组(P均<0.05),脂质体槲皮素组与5-氟尿嘧啶组比较差异无统计学意义(P均>0.05)。HE染色显示脂质体槲皮素组与5-氟尿嘧啶组肿瘤坏死区域较模型组小,2组病理表现相似。结论脂质体槲皮素对HT29实体瘤有较好的抑瘤作用。

Abstract:Objective It is to investigate the effect of liposome quercetin on ectopic solid tumors of human colon cancer adenocarcinoma cell line HT29 BALB/c in mice.Methods A tumor model of BALB/c mice of HT29 colon adenocarcinoma was established.48 model mice were randomly divided into model group,liposomal quercetin group,quercetin group,5-fluorouracil group,12 mice in each group.4 days after inoculation,the mice in the model group were injected intraperitoneally with 0.3 mL of normal saline,the ones in liposomal quercetin group was intraperitoneally injected with liposomal quercetin 50 mg/kg,and the ones in quercetin group was intraperitoneally injected with 50 mg/kg quercetin,5-fluorouracil group was injected intraperitoneally with 15 mg/kg 5-fluorouracil,they were injected once every 3 days and totally for 8 times.During the intervention,the tumor volume and body weight of the mice in each group were recorded;the mice of each group were sacrificed in 3 days after the last injection,the tumors were stripped,weighed,tumor volume was measured,tumor volume inhibition rate and tumor mass inhibition rate were calculated,and HE pathology was used to observe tumor pathology.Results No mice died in each group,and the tumor volume of liposomal quercetin group and quercetin group was significantly smaller than that of model group(P<0.05),but there was no significant difference in body weight between the groups(P>0.05);the tumor volume inhibition rates of the liposome quercetin group,quercetin group,and 5-fluorouracil group were 25.43%,22.27%,and 26.06%,respectively,and the tumor quality inhibition rates were 15.47%,13.37%and 16.66%.The rates in the liposomal quercetin group and the 5-fluorouracil group were significantly higher than those in the quercetin group(P<0.05).There was no significant difference between the liposomal quercetin group and the 5-fluorouracil group(P>0.05).HE staining showed that the tumor necrosis area in the liposome quercetin group and the 5-fluorouracil group was smaller than that in the model group,and the pathological manifestations of the two groups were similar.Conclusion Liposomal quercetin has a good antitumor effect on HT29 solid tumors.
