亚稀褶红菇染毒实验动物与中毒人群心肌毒性比较研究

摘    要:目的 为研究亚稀褶红菇中毒人群与染毒实验动物心肌损伤相关性。方法 采用发生过食物中毒原产地经鉴定为亚稀褶红菇的蘑菇标本,建立动物模型进行试验。设亚稀褶红菇625mg/kgBW、1250mg/kgBW、2500mg/kgBW剂量组和阴性对照组(等体积生理盐水),40只雌雄大鼠(180g~220g)按体重随机分为4组,染毒后第24h采血,测定Mb、cTnI、LDH、CK、CKMB、HBDH、ALT、AST。与人群流行病学资料的心肌酶谱比较,比较研究实验动物心肌毒性机制与人体流行病学毒性反应特征的相关性。结果 雌性SD大鼠高剂量组的血清生化指标MYO、cTnI、HBDH、CKMB 、CK、LDH、ALT、AST值与阴性对照组比较,明显增高,差异有统计学意义(P<0.05)。雄性SD大鼠高剂量组的血清生化指标cTnI、HBDH、CK、LDH、ALT、AST值与阴性对照组比较,明显增高,差异有统计学意义(P<0.05)。中剂量组雄性SD大鼠cTnI、雌性SD大鼠HBDH、LDH、AST值与阴性对照组比较,明显增高,差异有统计学意义(P<0.05)。其中cTnI指标升高,呈剂量反应关系。人群流行病学报告中心肌酶谱指标标MYO、CKMB 、CK、ALT、AST值显著高于正常值,患者死亡率达75%。结论 动物试验的结果与人群流行病学报告相吻合,亚稀褶红菇中毒对生物体的心肌功能具有损害。

Abstract:To study the correlation between myocardial damage in the poisoned people of theRussula subnigricansHongo and the exposed experimental animals.Methods Animal specimens of theRussula subnigricans Hongo,which hadoriginated in food poisoning,were used to establish an animal model for testing.Set 625 mg/kg BW,1 250 mg/kg BW,2 500mg/kg BW dose group and negative control group(equal volume of normal saline),40 rats were randomly divided into 4groups according to body weight,and blood was collected at 24 h after exposure.Mb,c Tn I,LDH,CK,CKMB,HBDH,ALT,AST.Compared with the myocardial zymogram of population epidemiological data,the relationship between the myocardialtoxicity mechanism of experimental animals and the characteristics of human epidemiological toxicity was compared.ResultsCompared with the negative control group,the serum biochemical indexes myo,c Tn I,HBDH,CKMB,CK,LDH,alt,AST inthe high dose group were significantly higher(P<0.05).The serum biochemical indexes c Tn I,HBDH,CK,LDH,alt,ASTin the high dose group of male SD rats were significantly higher than those in the negative control group(P<0.05).Thevalues of c Tn I,HBDH,LDH and AST in male SD rats and female SD rats in the middle dose group were significantly higherthan those in the negative control group(P<0.05).CTn I increased in a dose-response manner.In the epidemiologicalreport,the MXY,CKMB,CK,ALT,and AST values of the myocardial enzyme index were significantly higher than the normalvalue,and the patient mortality rate was 75%.Conclusion The results of animal experiments are consistent with theepidemiological report of the population.The poisoning of theRussula subnigricans Hongo has impaired the myocardial functionof the organism.
