瑞舒伐他汀对颈动脉粥样硬化大鼠血管损伤及MMP-2、MMP-9和PKC表达的影响

摘    要:目的 探究瑞舒伐他汀对颈动脉粥样硬化大鼠血管损伤的保护作用,以及对基质金属蛋白酶-2(MMP-2),基质金属蛋白酶-9(MMP-9)和蛋白激酶C(PKC)表达的影响.方法 将90只大鼠随机分为对照组、模型组和瑞舒伐他汀组,每组30只,模型组和瑞舒伐他汀组给予高脂鼠粮并复制颈动脉球囊损伤模型.检测颈总动脉内膜、中膜厚度,RT-PCR法检测颈总动脉内MMP-2 mRNA、MMP-9 mRNA和PKC mRNA表达水平,Western blotting法检测颈总动脉内MMP-2蛋白、MMP-9蛋白和PKC蛋白表达水平.结果 瑞舒伐他汀组与模型组比较,精神恢复较快,毛发色泽光滑,活动量较大,体质量增加速度较快;瑞舒伐他汀组内膜厚度/中膜厚度比值、MMP-2 mRNA、MMP-9 mRNA、PKC mRNA、MMP-2蛋白、MMP-9蛋白和PKC蛋白表达水平与模型组比较,差异有统计学意义(P<0.001).结论 瑞舒伐他汀对动脉粥样硬化大鼠颈总动脉损伤有保护作用,保护作用机制可能为下调MMP-2,MMP-9和PKC表达水平.

Abstract:Objective To explore the effect of rosuvastatin on blood vessel damage of neck arteries and the expression level of MMP-2, MMP-9 and PKC in rats with carotid atherosclerosis. Methods Ninety rats were randomly assigned into control group,model group and rosuvastatin group,30 rats in each group. The rats in model group and rosuvastatin group were fed high-fat diet tomake carotid artery in-jury model. The intima-media thicknessof carotid arterywas detected. The expression levels of MMP-2 mRNA,MMP-9 mRNA and PKC mRNA in carotid arteryof rats were detected by RT-PCR. The expression levels of protein of MMP-2,MMP-9 and PKC in carotid artery of rats were detectedby Western blotting. Results Compared with model group,rosuvastatin group rats were characterized bythe spirit recover faster,hair color and smooth large amount of activity,weight gain faster. The ntima-media thickness of carotid artery and the ex-pression levels of MMP-2 mRNA,MMP-9 mRNA,PKC mRNA,MMP-2 protein,MMP-9 protein and PKC protein were significantly high-er than model group(P < 0. 001). Conclusions Rosuvastatin has the effect of protection in blood vessel damage of neck arteries. The mechanismof rosuvastatin might through reducing the expression levels of MMP-2,MMP-9 and PKC.
