蝮蛇抗栓酶对离体工作员心缺血再灌注损伤的保护作用

摘    要:本实验采用Langendorff离体心脏灌流模型,观察了大鼠心肌缺血(15cm H_2O,20 min)再灌注(70cm H_2O,30 min)后心肌的机能、代谢改变及蝮蛇抗栓酶对其影响。实验分为单纯缺血再灌注组(I-R组)和蝮蛇抗栓酶治疗组(A.H组,灌流液中加入A.H 2.5 u/L)。结果发现,A.H组心肌游离脂肪酸(FFA)降低,磷脂(PL)含量高于I-R组;心肌脂质过氧化物(LPO)含量降低,锰-超氧化物歧化酶(Mn-SOD)及谷胱甘肽过氧化物酶(GSH-Px)活力高于I-R组。结果提示,A.H在体外具有较强的抗自由基损伤、保护抗氧化酶活性和稳定细胞膜的作用。

Abstract:The functional and metabolic changes in isolated working rat heart following myocardial ischemia (15cm H_2O, 20min) and reperfusion (70cm H_2O, 30min), and the effect of A.Halys on the above changes were observed in the Langendorff Heart Model. The study was divided into two groups: ischemia - reperfusion group (I- R group) and A.Halys-treated group (A.H group, with addition of A.Halys 2.5U/L).The results showed there were decreases in FFA, MDA contents and increases in phospholipid contents, and Mn-SOD and GSH-Px activities in the A.H group, as compared with the 1-R group. The results suggested that A.Halys had a beneficial effect on ischemic myocardium, as indicated by tle inactivation of lipid peroxides, and an increase of antioxidant enzyme activities and the stabilization of cell membrane.
