摘    要:用液闪计数测定经组织培养的6株人肿瘤(MGC 80-3、SGC-7901、BEL-7402、Os-732、Detroit-6和 HeLa)细胞及1株正常成纤维细胞对~3H-HPD 的摄取和存留。MGC 80-3和成纤维细胞的摄取量,在测定的24h 内,随温育时间延长而增加;温育30 min 换液,再培养30 min,血卟啉衍生物含量就明显减少,MGC 80-3和成纤维细胞分别只含原摄入量的32.01%和40.75%;但在以后的24h 内,维持在类似的水平上。7株细胞温育30min 摄入~3H-HPD 量,按多少顺序依次为成纤维细胞、HeLa、BEL-7402、Detroit-6、SGC-7901、MGC 80-3 及 Os-732;但是,培养24h 后,由于各株细胞排出量不同,~3H-HPD 存留率大小顺序为 Os-732、Detroit-6、BEL-7402、HeLa、SGC-7901,MGC 80-3 及成纤维细胞。环境因素对细胞摄取~3H-HPD 起重要作用,提高 pH 值、加入血清或降低温度都使摄入减少。本文结果提示,恶性组织比正常组织血卟啉衍生物含量高,不是因为恶性细胞摄入多,而是由于存留时间长,与体内实验结果一致,可供 HPD 光敏治疗恶性肿瘤作参考。

Abstract:The uptake and retention of ~3H-HPD in.human malignant cells(cell line MGC 80-3,SGC-7901,BEL-7402,Os-732,Detroit-6 and HeLa)and normal fibroblasts were studied by liquid scintillation counting method.The order of cell lines which took up ~3H-HPD after 30 min.incubation were fibroblasts,HeLa, BEL-7402,Detroit-6,SGC-7901,MGC 80-3 and Os-732 respectively according to the amount of ~3H-HPD in cells.Due to the variety in the amount of excretion, however,those cells which retained ~3H-HPD after 24 hours cultivation were Os-732, Detroit-6, BEL-7402,HeLa,SGC-7901,MGC 80-3 and fibroblasts respectively in respect to the ~3H-HPD level in cells.The uptake was reduced by raising extracel- lular pH and lowering temperature as well as adding higher concentration of serum. Our results showed that the ~3H-HPD content in malignant ceils was higher than those in normal ones and it was because that malignant cells retained ~3H-HPD longer other than took up more
