自体骨髓细胞局部移植对外周血管新生作用的影响

摘    要:目的:探讨自体骨髓细胞局部移植对肢体缺血的治疗作用。方法: 14只雄性新西兰兔结扎并离断单侧股动脉及其分支动脉1 h后,骨髓细胞局部移植组(n=7)于缺血局部肌肉注射新鲜分离的自体骨髓单个核细胞悬液,对照组(n=7)局部肌肉注射磷酸盐缓冲液,分别于实验前(0 d)、7 d和28 d进行股部血流动力学检测,于28 d时处死动物做组织病理学分析。结果: 7 d和28 d时,彩色多谱勒血流显像示骨髓细胞局部移植组血流信号较对照组丰富,且收缩期峰值血流速度与舒张期流速较快(P<0.05),28 d时血管阻力指数、血流量两组间有显著差异(P<0.05);病理学分析骨髓细胞局部移植组单位面积毛细血管密度显著高于对照组(P<0.01),毛细血管数目与肌纤维数目的比值两组间显著差异(P<0.05)。 结论: 自体骨髓细胞局部移植促进外周血管新生,可能成为缺血性外周血管疾病一种新的治疗方法。

Abstract:AIM: To investigate the effect of autologous bone marrow mononuclear cells (BM-MNCs) local transplantation (BM-LT) on a rabbit model of hindlimb ischemia. METHODS: After unilateral femoral artery and its branches were surgically ligated and excised in rabbits (n=14) 1 hour or so, autologous BM-MNCs were injected into the ischemic skeletal muscles in 7 rabbits, and phosphate buffered saline for another 7 ones as control. Hemodynamics parameters were measured at day 0, day 7 and day 28, respectively. On day 28, the animals were sacrificed in groups for pathological analysis. RESULTS: Compared with control group, color Doppler flow image in BM-LT group was recovered significantly, and its' systolic peak velocity and end-diastolic velocity were also improved (P<0.05). Resistance index and flow volume were significantly improved on day 28 (P<0.05). A higher capillary density was detected in BM-LT group than that in control group (P<0.01). Ratio of the number of capillary and the number of myocyte fiber were significant difference (P<0.05). CONCLUSION: BM-LT improves peripheral neovascularization. It might provide a new therapeutic method for ischemic peripheral vascular disease. [
