人碱性成纤维细胞生长因子基因真核表达载体的构建

摘    要:目的:构建人碱性成纤维细胞生长因子bFGF基因真核表达载体,探讨bFGF基因转移治疗视网膜病变的方法.方法:将bFGF和GFP基因克隆到真核表达载体pcDNA3中.结果:酶切及琼脂糖电泳分析 pcFG中含有bFGF和GFP基因.结论:成功的构建了含有绿色荧光蛋白的人碱性成纤维细胞生长因子基因的真核表达载体.

Abstract:Objective: To construct eukaryotic expression plasmid of human basic fibroblast growth factor gene (hbFGF) and green fluorescent protein (GFP) for further study on bFGF gene therapy of retinitis pigmentosa. Methods: The hbFGF and GFP genes were inserted into the eukaryotic expression plasmid pcDNA3. Results: The pcFG plasmid was confirmed to containe correct nucleotide sequence for hbFGF and GFP gene by restriction endounclease digestion and agarose gel electrophoresis. Conclusion: A eukaryotic expression plasmid containing hbFGF and GFP gene is successfully constructed in the study.
