Bcl-2、bax、P53蛋白表达与老年妇女外阴营养不良细胞凋亡的相关性研究

目的 :探讨老年妇女外阴营养不良组织中细胞凋亡及相关基因的表达情况。方法 :采用原位末端标记及免疫组化S -P法检测 78例老年妇女外阴营养不良和 12例老年妇女外阴营养不良恶变石蜡包埋组织中的细胞凋亡及相关基因bcl- 2、bax、p5 3的表达情况。另取石蜡包埋的老年妇女正常外阴皮肤 10例作为对照。结果 :老年妇女正常外阴皮肤、外阴营养不良及外阴营养不良恶变中均存在细胞凋亡及bcl- 2、bax和p5 3的表达 ,细胞凋亡指数 (AI)及bcl- 2和bax的表达在营养不良组明显高于营养不良恶变组 (P >0 .0 1) ,而 p5 3的表达却相反 (P >0 .0 1)。在老年妇女正常外阴皮肤及外阴营养不良中凋亡细胞的分布区域与表达bcl- 2和bax蛋白的细胞分布区域一致 ,bcl- 2与bax表达呈正相关 (P >0 .0 5 ) ;在恶变部位凋亡细胞的分布区域与突变型 p5 3蛋白表达呈负相关 ,p5 3分别与bcl- 2和bax的表达呈负相关 (P >0 .0 5 )。结论 :老年妇女外阴营养不良的发生发展和恶变与细胞凋亡有关 ,bcl- 2、bax和p5 3起一定作用。

ASSOCIATION BETWEEN EXPRESSIONS OF BCL-2,BAX ,P53 AND APOPTOSIS IN THE VULVAR DYSTROPHY OF ELDERLY WOMEN

Objective:To investigate the relationship between apoptosis and the expression of bcl-2, bax and p53 in vulvar dystrophy of the elder women.Methods:Apoptosis index and the expression of bcl-2, bax and p53 were determined with in sim end labelled assay and immunohistochemical technique, in 78 specimens of vulvar dystrophy ,12 of neoplastic vulvar dystrophy ( turned into vulvar squamous cell carcinoma ),and 10 of normal vulvar skin served as controls.Results:Apoptosis and the expression of bcl-2,bax p53 were presented in vulvar dystrophy tissues. Apoptosis index and the expression of bcl-2 and bax increased significantly in vulvar dystrophy than those in the neoplastic vulvar dystrophy ( P <0.01), but the rate of p53 expression reduced. In normal vulvar skin and vulvar dystrophy, the distribution of the apoptotic cells was identical with that of bcl-2 and bax ,and there was a positive correlation between the expression of bcl-2 and bax( P <0.05). But in neoplastic vulvar dystrophy,there was a negative correlation between the distribution of the apoptotic cells with the expression of mutated p53, and also a negative correlation between the expression of p53 and bcl-2 or p53 and bax(( P <0.05).Conclusions:The development of vulvar dystrophy may be related with apoptosis. The related apoptic genes, bcl-2 ,bax and p53 may play a role.
