生存素反义寡核苷酸对人胃癌细胞增殖的抑制作用

摘    要:目的 探讨生存素(survivin)反义寡核苷酸(ASODN)对人胃癌细胞SGC7901增殖的抑制作用。方法 设计合成特异性靶向生存素的反义寡核苷酸。胃癌细胞株SGC7901分为4组:空白对照组、单纯脂质体对照组、正义链转染对照组、ASODN转染组。作用48h后收获各组细胞。Westernblot法检测各组细胞生存素表达情况,流式细胞仪检测各组细胞凋亡率,MTT法检测各组细胞的生长抑制率。结果 脂质体介导生存素反义寡核苷酸转染后的胃癌细胞出现生存素蛋白表达明显下降;ASODN转染组细胞凋亡率明显高于各对照组(P〈0.05),而各对照组问差异无显著性(P〉0.05);ASODN转染组细胞的抑制率明显高于各对照组(P〈0.05),而各对照组问差异无显著性(P〉0.05)。结论 生存素反义寡核苷酸转染胃癌细胞能下凋生存素蛋白表达,诱导胃癌细胞凋亡,抑制细胞增殖,具有明显的抗癌作用。

Abstract:Objective To investigate the inhibitory effects of survivin antisense oligonucleotide(ASODN) on human gastric carcinoma cell line SGC7901.Methods ASODN targeting survivin was designed and constructed.Cultured cancer cells were divided into 4 groups: sham control group,lipofectin control group,oligonucleotide(SODN) control group and ASODN test group.After transfection for 48 h,cultured cells were harvested to proceed with the tests for: survivin protein expression by Western blot,apoptotic rate(AR) by flow cytometry,the rate of inhibition(IR) by colorimetric MTT cell viability/proliferation assay.Results The expression of survivin in SGC7901 cancer cells decreased after being transfected with survivin ASODN.The AR and IR in ASODN test group were significantly higher than in the control groups.Conclusions The expression of survivin may be decreased in gastric cancer cells after ASODN transfection.Survivin ASODN can induce apoptosis and inhibit proliferation of SGC7901 cells,and hence,may well be a prospective anti-cancer drug.
