摘    要:目的:研究针刺对大鼠学习记忆的影响,分析针刺影响大鼠学习记忆与其影响脑内自由基反应的关系。方法:筛选体质量和灵活性等较一致健康大鼠40只,按随机数字表随机分成4组,每组10只。其中针刺组用30#美容针于实验开始之日起针刺固定于自制大鼠固定架上的大鼠肾俞、百会穴。百会向前平刺3~5mm,肾俞向内斜刺5mm,接G6805电针仪刺激10min,连续波,频率为3Hz,以大鼠肢体抖动为刺激强度标准,1次/d,连续7d;尼莫组灌服尼莫地平混悬液10mL/kg,1次/d,连续7d;模型组和空白组自实验第1天起,与针刺组同样方式固定,10min/次,1次/d,连续7d。采用水迷宫测试正常大鼠和亚硝酸钠所致痴呆大鼠的学习记忆能力,同时测定被试大鼠脑内过氧化脂质(LPO)含量和超氧化歧化酶(SOD)活性。结果:在第5天训练末,针刺组在(9.3±2.6)s即可登陆,而空白组在(45.5±7.6)s才能成功登陆,说明针刺能提高正常大鼠的学习记忆能力;造模后模型组登陆时间持续居高,而针刺组登陆时间逐渐缩短,较之模型组差异有显著性意义(t=2.982,P<0.05)。此外,在第12天的末次测试中,模型组错误次数为(5.71±1.37)次,针刺组分别为(0.73±0.25)次,均较模型组减少,差异具有非常显著性意义(t=3.576,P<0.01),提示针刺能拮抗亚硝酸钠对大鼠空间识别记忆巩固的破坏;针刺组大鼠

Abstract:AIM:To investigate the effect of acupuncture on the learning and memory of rats and analyze the relationship between the effect and reaction of free radicals. METHODS:Forty healthy rats with similar body mass and flexibility were randomly divided by number table into 4 groups with 10 rats in each group.In acupuncture group,cosmetological needles were punctured into Shenshu and Paihui of rats which were fixed on self made fixator.Needles were punctured forward and horizontally into Paihui for 3- 5 mm while inward and obliquely into Shenshu for 5 mm.G6805 electric stimulator was connected with continuous wave,frequency of 3 Hz.With shaking of limbs as intensity standard,rats were stimulated once a day for 7 days;Nimodipine group,nimodipine suspension,10 mL/kg,was injected once a day for 7 days.Animals in model group and blank group were fixed with same method from 1 day,10 min each time,once a day and for 7 days.Water maze was used to test the learning memory of normal rats and dementia rats induced by sodium nitrite. The content of brain Lipoperoxide(LPO) and the activity of Superoxide dismutase(SOD) of the rats were also determined. RESULTS:At 5 day,rats in acupuncture could land at (9.3±2.6) s while in blank group,it was (45.5±7.6) s,suggesting that acupuncture could improve the learning memory of normal rats.The landing time was kept in a longer time in model group while the landing time was gradually shortened in acupuncture group(t=2.982, P< 0.05).Besides,in 12d test,the mistake times was (5.71±1.37) while in acupuncture group,it was (0.73±0.25). The difference was significant(t=3.576,P<0.01),suggesting that acupuncture can antagonize the destruction of space learning and memory consolidation by soldium nitrite. In the acupuncture group, The content of LPO [(15.4±0.4) μ mol/g] in the brain cortex was lower than that [(17.2±0.6) μ mol/g] in the model group, but the SOD activity[(101.9±2.2) pmol/(s@g)] was higher than that[(70.4±1.8) pmol/(s@ g)] in the model group (P< 0.05 or P< 0.01).SOD/LPO was negatively correlated to learning ability. CONCLUSION:There is some relationship between the effect of acupuncture in promoting the learning of rats and its effect of inhibiting brain free radical reaction.
