三氧化二砷诱导血管内皮细胞凋亡和抑制血管生成的研究

摘    要:目的:研究三氧化二砷(As2O3)抑制血管内皮细胞(HUVECs)生长、诱导凋亡和抑制血管生成的作用。方法:As2O3与HUVECs共同孵育一定时间后,观察细胞形态学变化,并用MTT方法测定细胞生长曲线图、TUNEL方法检测细胞凋亡;观测As2O3对鸡胚血管生成的影响。结果:在0.75μmol/L~6μmol/L浓度范围内随药物浓度增加或作用时间延长,细胞生长抑制和细胞凋亡效果越显著;As2O3作用鸡胚绒毛尿囊膜后血管生成减少。结论:As2O3抑制HUVECs生长、诱导细胞凋亡,对血管生成有抑制作用。

Abstract:Objective: To study the effect of arsenic trioxide (As2O3) on human vascular endothelium cells (HUEVCs) and angiogenesis. Methods: As2O3 and HUEVCs were incubated together for some time to observe the cytomorphological changes. To observe the effect of As2O3 On the angiogenesis of chick - embryo vascular, MTT reduction assay was used to detect the cytogrowth curve and TUNEL was employed to measure the cell proliferation. Results: In the concentration range of 0.75 - 6 umol/L, As2O3 - induced cell apopto-sis and As2O3 - inhibited angiogenesis were enhanced in time - and - concentration - dependent manner. What' s more, As2O3 significantly inhibited the angiogenesis in chorio - allantioc membrane (CAM). Conclusion: As2O3 can inhibit the growth of HUEVCs, induce the apoptosis of cells and reduce the angiogenesis in CAM.
