外源性NGF对家兔急性全脑缺血再灌注期间兴奋性氨基酸释放的影响

摘    要:目的:观察外源性神经生长因子(NGF)对家兔急性全脑缺血再灌注期间兴奋性氨基酸释放及家兔生存情况的影响,以探讨NGF对脑神经细胞的保护机制。方法:用反相高效液相色谱(HPLC)荧光检测方法,测量急性全脑缺血再灌注损伤模型动物脑脊液中的氨基酸,特别是兴奋性氨基酸含量。并观察实验动物生存情况。结果:脑缺血再灌注期间,脑脊液中兴奋性氨基酸(EAA)明显增加(P<0.01);而外源性NGF可使上述指标明显下降,与对照组有明显差异(P<0.05和P<0.01),结论:外源性NGF可能过血脑屏障,抑制脑内EAA过度释放,改善缺血再灌注对脑的损害,保护脑细胞,提高动物生存率,在临床脑复苏中有一定应用前景和实用价值。

Abstract:Objective To observe the effects of exogenetic NGF in rabbits with acute whole cerebral ischemia and reperfusion in releaving excitatory amino acid and on survival condition,to evaluate NGF on the protective mechanism of cerebral cell.Methods High perfusion liquid chromatography(HPLC),fluorimetry was used to determine the content of amino acid,especially the content of excitatory amino acid in cerebrospinal fluid of experimental animals of acute whole cerebral ischemia animals of acute whole cerebral ischemia and reperfusion and the survival condition of experimental animals was observed.Results During cerebral ischemia and reperfusion,the content of the excitatory amino acid(EAA)in cerebrospinal fluid was remarkably increased( P <0.01),while the exogenetic NGF made the abore indications evidently reduceel,which were evidently different from the control group( P <0 05 and P <0 01).Conclusion Exogenetic can pass blood-brain barrier inhibit intracerebral EAA from over-releaving,which could improve cerebral injury by ischemia and reperfusion,protect brain cells and increase animal survival rate.It shows that this experiment has certain promise and practical value in clinical brain anobiosis.
