| 摘 要： | 目的:建立人体血浆中瑞芬太尼(Remifentanil,REF)的高效液相色谱/质谱串联(HPLC-MS/MS)的分析方法。方法:血浆中REF用乙酸乙酯作液-液提取,以高效液相色谱-质谱联用技术法测定REF血药浓度。结果:血浆中瑞芬太尼浓度在0.1～25ug.L-1范围内呈良好的线性关系,典型代表方程为:Y=0.2690*X-2.9549*10-4,r2=0.9944(n=7),最低检测浓度为0.1ug.L-1(S/N>10)。REF血浆样品的低、中、高三个浓度(0.2ug.L-1、1.25ug.L-1和12.5ug.L-1)其日内、日间变异系数(RSD)均不大于±10%,其方法回收率均不低于80%。结论:该法操作简便、结果准确,可用于REF儿科临床应用和药动学研究的血药浓度测定。

| Abstract: | Objective. To set up a HPLC-MS/MS method for determination of the blood concentrations of Remifentanil（REF）. Methods. REF was extracted from the plasmas with ethyl acetate and analysed on HPLC-MS/MS. Results. The liner ranges of REF was 62. 5-1000 ug. L-1 （r2 =0. 9972）. The limit of quan- tity was 62. 5 ug. L-1. The RSD within-day and between-days were less than ± 10%. The rates of recovery absolutely was more than 80%. Conclusion. The method was shown to be simple, convenient, accurate and suitable for monitoring the blood concentrations of REF. |
